Objective To investigate the expression of tumor suppressor gene gene p21 and p27 in hepatocellular carcinoma(HCC) in whole blood of the patients with the expression level and the relationships between those two genes, To explore the relationship between the expression level of p21 and p27 in whole blood and the clinical data of patients with HCC.To provide evidence for screening and one or two prevention of HCC at high risk groups.Methods In this study, patients with HCC were used as the case group, Healthy people who were admitted to the hospital for medical examination were used as the control group., Collection of patients and control group of age, gender and other general information, HBV infection history, drinking history, smoking history, diet habit, education level, occupation and so on. Collection of patients with tumor details, including: tumor size, number, whether there is a portal vein tumor thrombus, cirrhosis, portal hypertension, and so on. Peripheral blood samples from study subjects were used as samples., total RNA extracted from whole blood, Reverse transcription of RNA to form the first chain c DNA, using c DNA as the template, with GAPDH as an internal control for PCR amplification and application of q PCR(quantitative real time PCR) method of PCR process for real-time detection, to understand the relative expression quantity of sample in the p21 gene and p27 gene, and to analyze the correlation of the two, also respectively compared the two gene relative expression quantity with hepatocellular carcinoma in patients with clinical data correlation. Using SPSS13.0 statistical software for data processing, count data using chi square test, measurement data using mean + standard deviation, the normality and homogeneity of variance test data are consistent with positive normal distribution and equal variance, so the use of independent samples t test. The relative expression of p21 gene and p27 gene was analyzed by Pearson correlation. P<0.05 difference was statistically significant. Results P21 m RNA expression level was higher in the hepatocellular carcinoma patients than in the not tumor patient(P < 0.05). p21 gene in case group of relative expression quantity is higher than the control group, the difference is statistically significant(P < 0.05). Through cases clinical data and p21 gene relative expression is known, drinking, history of hepatitis, preoperative AFP value and p21 gene relative expression quantity, the difference was statistically significant(P < 0.05) and p21 gene relative expression has no related to age, gender, smoking, portal high pressure, tumor number, tumor size, capsule, cirrhosis of the liver, with portal vein tumor thrombus(P > 0.05). P27 m RNA expression level was higher in the hepatocellular carcinoma patients than in the not tumor patient(P < 0.05), p27 gene in HCC group relative expression quantity is higher than the control group, the difference is statistically significant(P < 0.05); p27 gene relative expression quantity and cases clinical data shows, p27 gene relative expression has no relation with age, sex, alcohol, smoking, history of hepatitis, preoperative AFP value, portal high pressure, tumor number, tumor size, capsule, cirrhosis of the liver, with portal vein tumor thrombus(P > 0.05). Case group and the control group in the p21 gene and p27 gene m RNA relative expression amount of correlation showed that case group(r=-0.44) showed a negative correlation, control and there is a positive correlation between the group(r=0.197), but the correlation coefficients were close to 0, so the two genes have little relationship,needs to increase the amount of repeated experiments confirmed. Conclusion:(1) the expression of p21 and p27 gene in patients with hepatocellular carcinoma(HCC) in peripheral blood levels, suggesting that the expression level of p21 and p27 gene may be associated with HCC occurrence and development and to detect the blood expression level is expected to be early indicators of liver cancer.(2) Drinking, hepatitis B history, preoperative AFP level can make the m RNA gene p21 expression level increased.
