Hepatocellular carcinoma (HCC) was one of the common malignant tumors in humans.With a high incidence,poor treatment and high mortality, many patients were found when the tumor already had been in the middle and late stages, and they lost the Optimal treatment period. According to the statistics, global annual number of deaths due to hepatocellular carcinoma had been as high as59.8million, that’s to say,the number of deaths was third role in all deaths due to malignant tumors caused all over the world.The reasons of poor prognosis about HCC were involved to high degree of malignancy, the rapid progress of the disease and early metastasis so on. The treatment of HCC was mainly surgical treatment, and supplemented by interventional chemoembolization and immunosuppressants treatment.However, the effect was still not satisfactory.The Extracellular signal-regulated kinase(ERK) was an important component part of Mitogen-activated protein kinases(MAPKs) signal transduction system.The ERK was mainly composed of ERK1and ERK2, and it was one of the important members of MAPKs family. For the past few years, the study had found that ERK1/2pathway played an important significant biological effects in tumorigenesis, development, invasion, metastasis, and many other aspects. The tumor occurrence and development associated with the ERK1/2abnormal and (or) persistent activation. Siah2was an E3ubiquitin ligase,and there were two different components in mammal,Siahl and Siah2,which participated in a variety of physiological activities of the cell. It was currently confirmed that the abnormal expression of Siah2could promote the formation of tumor, and Siah2had a closely relationship with progression and metastasis of tumor. It was reported that ERK signal transduction pathway and Siah2protein had some contactions in occurrence and developmence of tumor.However,it was not know that how the Siah2played a role in tumor according to ERK1/2. The main subject was detected the expression of Siah2and ERKl/2in hepatocellular carcinoma and studied the preliminary exploration of the correlation between Siah2and ERK1/2.ObjectiveIn this issue, we used the technology of Immunohistochemistry and Western blotting to explore the expression of Siah2and ERKl/2in Hepatocellular carcinoma (HCC),and investigated their correlation in growth and development of HCC and its clinical significance.Methods1. The expression of Siah2and ERK1/2was detected by Immunohistochemistry, and there were49cases of HCC,34cases of non-tumor tissues and15cases of normal liver tissues.2. The expression of Siah2and ERK1/2in which49cases of HCC、34cases of non-tumor tissues and15cases of normal liver tissues was detected by western blotting.3. To compare the expression of Siah2and ERK1/2in cases of HCC,cases of non-tumor tissues and cases of normal liver tissues, and to investigate their correlation in growth and development of HCC. Results1. With the results of Immunohistochemistry, there was significance in the expression levels of Siah2and ERK1/2in HCC tissues,non-tumor tissues and normal liver tissue (P<0.05). In HCC tissues, The positive rate of expression were significantly higher than that in the non-tumor tissues (P<0.05) and normal liver tissue (P<0.05).2. With the results of western blotting, there was significanty difference in the expression levels of Siah2and ERK1/2in HCC tissues,non-tumor tissues and normal liver tissue (P<0.05), and The expression level of Siah2and ERK1/2in HCC tissues was higher than that in non-tumor tissues and normal liver tissues(P<0.05).3. With the results of application of statistical analysis software, the expression of Siah2in HCC was positively correlated with that of ERK1/2(P<0.05),and the expression of Siah2in HCC may be associated with ERK1/2.ConclusionSiah2and ERK1/2were over-expressive in HCC tissues,and The expression level of Siah2and ERK1/2in HCC tissues was higher than that in non-tumor tissues and normal liver tissues. The level expression of Siah2and ERK1/2was related to the degree of tumor differentiation and tumor size.The expression of Siah2and ERK1/2in the HCC may had some some connection.
