ObjectiveResently, researches have shown that klotho gene is closely related to a varietyof symptoms of aging. This study compared to assess import klotho gene and notimport the klotho gene of two groups of diabetic rats, and coronary artery lesiondegree and the change of correlatively biochemical index associated artery disease.Meanwhile, we explore klotho gene expression and the degree of the coronary arterylesions in rats and closely related to coronary artery lesions of the correlation betweenthe biochemical indexes.MethodsAccording to the Klotho gene sequence primers, applicating the PCRamplification purpose gene to build Klotho carrier and establishing diabetic ratsmodel. The satiety rats of blood glucose greater than16.7mmol/L were diabetesstandards. The diabetic rats were randomly divided into three groups, three groupswere diabetic model group, diabetic model and viral transfection group (treatmentgroup), diabetic model and negative carrier group (control group), respectively. Themodel group without treatment, the treatment group was given a one-time intravenousadenovirus Ad-rklotho, and the control group only received an injection Ad-EGFP.After12weeks, the animals pattern were sacrificed, and detecting each group theblood low-density lipoprotein, high-density lipoprotein, Choice of P elements andnitric oxide level, Coronary artery intima thickness, Lining in the film thickness ratio,the expression of Nitric oxide synthase (Nos). The Klotho gene mRNA and proteinexpression level were measured, and determining genes into effect. Results1.The rats of treatment group, model group, negative control group between age,body weight and blood glucose levels had no statistical significance (P>0.05).2.The klotho gene of treatment group mRNA and protein expression levels are obviously higher than model group and negative control group (P<0.05),which indicates that the import of Klotho adenovirus is ideal and the Experimental model meets the requirements.3.The coronary artery intima thicknessand lining in the film thickness ratio oftreatment group (1.740±0.002) rats have distinctly preponderance compared to the model group(2.150±0.010) and negative control group(0.260±0.004), and there is statistical significance(P<0.05).4.The biochemical criterion: the high-density lipoprotein of treatment group rats(0.780±0.026) is higher than the model group(0.581±0.001) and negative control group(0.618±0.001);the low-density lipoprotein of treatment group rats(0.352±0.001) with the model group(0.2±0.004) and negative control group has no significant difference. There are certain advantages of Nos with the express treatment group compared to model group and negative control group, so no statistical difference(P <0.05); the treatment group rats P-select element has a higher standard with themodel group and the negative control group(P<0.05);While the indicators data between model group and the control group has no statistical significance (P>0.05).ConclusionsThe mRNA of Klotho gene and protein expression of The treatment group ratswhich with Adenovirus packaging Klotho gene have a higher level than the modelgroup and the negative control group. The high-density lipoprotein, P-selectelement, Nos et al. and Chronic coronary artery lesions related biochemicalindicators in the treatment group rats compare with the model group and the negativecontrol group has Statistically significant. The coronary artery intima thickness and lining in the film thickness ratio of treatment group rats have distinctly preponderancecompared with the model group and negative control group.Klotho gene overexpression can affect the level of the high-density lipoprotein,P-select element and Nos, in addition, the overexpression has a definite correlationwith Coronary lesions of diabetic rats. That is to say, it can slow down the process ofthe coronary artery lesions of diabetic rats, and it is a Influence Index of coronaryartery lesions.
