Objective:This study makes EAE models by wistar rats to observe that Zi-yin-gu-ben-fanggrades EAE rats’behavior、weight and neurological function and the pathological change ofthe lateral ventricle tissues of rats and MBP gray number fusion expression on it.It exploresthis Chinese medicine prescription’s mechanism of action about EAE rats’brain tissues tomake new experimental theoretical basis of the ZI-yin-gu-ben-fang’s clinical application onEAE、MS.Method:1.Give the spinal cord homogenate of fresh guinea pigs adding Complete Freund’s Adjuvant,give GPSCH-CFA0.1ml intracutaneous injection in the four pad limbs of EAErats as immunogen,give pertussis vaccine0.1ml intracutaneous injection in the rats’ hindlegs’ dorsum of feet to increase the permeability of the blood-CSF barrier to make modelsof rats.2.Divide76female wistar rats into6groups randomly:Model group16rats(EAE withnormal saline),hormone group (EAE with Dexamethasone Acetate tablets6.25mg/Kg.d),TCM high dose group (EAE with Chinese medicine53.3g/Kg.d),TCM middle dosegroup(EAE with Chinese medicine26.7g/Kg.d), TCM low dose group (EAE with Chinesemedicine13.3g/Kg.d),and blank group(treated with nothing),12rats each group.At the firstday we inject the immunizing antigen into all rats,and give Chinese medicine to every doseof TCM group7days before making models.However,we begin to give medicine to thehormone group at the second day.Give medicine each time a day.Then we should observe allgroup rats`neurological scores and the change of food consumption、weight and behavior ofthe rats.3.At the fifteenth day we take out the neurological tissue of the rats`the lateral ventricle tostain with hematoxylin and eosin and observe its pathology.And we should detect the MBPgray degree and compare the MBP expression of each group. 2.Results:1. Each group of rats behavior change:(1) the incidence of a disease: the blankgroup except for accidental death, incidence, download the index weight, nerve functionscore is zero; Compared with model group, Chinese medicine high, medium, low andhormone group was lower than those of model group,(P <0.05); Traditional Chinesemedicine (TCM) of high, middle dose group compared to the hormone group, no significantdifference (P>0.05); In the traditional Chinese medicine (TCM), low dose group is higherthan the hormone group (P <0.05); Middle and low dose group is higher than the high dosegroup (P <0.05).(2) the weights are: building before each weight of rats was used as thedifference is not obvious (P>0.05);15days after the building (acute phase), compared withmodel group, high, medium and low dose group, hormone group rats weight loss index islower than the model group (P <0.05); Compared with hormone group, Chinese medicinehigh, medium weight had no significant difference (P>0.05); High, medium and hormonegroup was lower than those of traditional Chinese medicine (TCM) to lower dose group (P <0.05).(3) nerve function score comparison:15days after the immunization, compared withmodel group, the hormone group, Chinese medicine high, medium and low dose group ofnerve function score were lower than that of model group (P <0.05); Traditional Chinesemedicine (TCM) compared with model group, low dose group nerve function scoredifference is not obvious (P>0.05); Traditional Chinese medicine (TCM) of high, middledose group, hormone, nerve function score had no significant difference (P>0.05).2.The pathological morphology change of each group:After immunizing15days,theinflammatory focus of model group is more than the other groups,they have significantdifference(p<0.05); the inflammatory focus of the hormone group is less than the middle、low TCM dose group(p<0.05); but there is no obvious difference between the hormonegroup and the high TCM dose group(p>0.05);so do the middle TCM dose group and theaverage group as well as the low TCM dose and the average group(p>0.05).3.The immunohistochemistry change of each group:The MBP fusion expression of thelateral ventricle around in rats of blank group is normal;but the model group is reducedobviously(p<0.01).The MBP fusion expression in rats of the high TCM dose group and thehormone group is more enhanced than the EAE,low and middle TCM group(p<0.01). Thereis no obvious difference between the high TCM dose group and the hormonegroup(p>0.05).The MBP fusion expression in rats of the high TCM dose is more enhanced than the low and middle TCM dose group(p<0.01).Conclusion:1.The EAE rats models copied in this study are successful,steady and reliable.2.ZI-yin-gu-ben-fang can decrease the incidence of EAE rats,the loss of rats’weight and theneurological scores during the illness period.It also can relieve clinical symptoms.3.ZI-yin-gu-ben-fang can relieve the degree of the inflammatory cell infiltration and edemain the central nervous system,reduce the immune response and protect the nerve injury inrats.4.ZI-yin-gu-ben-fang can obviously relieve the loss of MBP around the rats’ lateralventricle,reduce immune response and promote the regeneration and repair of MBP.
