Objective: The purpose of this paper is adopting transcriptomics method, the basicsyndromes of traditional Chinese medicine Qi and yin deficiency syndrome in patientswith HIV/AIDS as the object of study,compared with healthy people in the same area.Total RNA was extracted from peripheral blood, detected differences mRNA andmicroRNA of Qi and Yin deficiency syndrome with HIV/AIDS from healthy people.The quantitative PCR technique is used to confirm that the specific mRNA andmicroRNA of Qi and yin deficiency syndrome with HIV/AIDS, construction ofmolecular networks, finds transcriptomics material basis of the Qi and Yin deficiencywith HIV/AIDS, speculates signal of possible markers of Qi and yin deficiencysyndrome, plays an important role in evaluation on TCM syndrome type HIV/AIDSobjective research and curative effect. Using the classical prescription of Shengmaiyinfor drugs counterevidence, detects the prognosis of differential gene. To platform theestablishment of syndrome of transcriptome technology, in order to explore theessence of TCM syndrome to provide ideas and methods from the transcriptome level.Methods:Experimental samples are from AIDS patients with deficiency of Qi and Yintwo which are Contrasted with healthy people In some areas of Henan Province,Western diagnostic criteria use the national Ministry of health in2011edition of"AIDS treatment guidelines" diagnostic criteria. The Chinese medicine diagnosisstandard of the Syndrome of deficiency of Qi and Yin two use the Early stage ofthe project group’s the national project results "HIV/AIDS common syndromediagnostic scale". DNA chips which use Afffymetrix, applicate BRB-ArrayTools(Version4.2) software on a variety of gene chip data processing, use the singlevariable sample for variance test to choose the differental gene, analysisdown-regulation of gene ontology functions and information access, use DAVID Tools (http://david.abcc.ncifcrf.gov/) online analysis, analysis of gene interaction.Itmainly applicate STRING (http://string.embl.de) and BRB-ArrayTools4.2softwarewith transcription factors and related literature to analysis, clustering analysis usingstratification cluster, for each chip signal values genes is the center, metric centeredcorrelation, linkage two-way stratification cluster the sample and the differencesignal for average linkage.Results: The BRB-ArrayTools (Version4.2) software is used to analysis of variancefor the gene of patient group and healthy control group, data filtering is Percent ofdata missing or filtered out exceeds50%, Percent of absent (i.e., Detection, Call=A)data exceeds50%standard filter: accord with p<0.001; FDR<0.05, finds thedifferent mRNA739and the different MicroRNA177. Analysis of the relationshipbetween mRNA and microRNA: finds mRNA-MicroRNA153,96target genes,according to the interaction between the target gene, consisting of153nodes,260sides of the relationship graph. The96target genes involved in response to trauma,cell proliferation and apoptosis, cells to oxidative stress,GNAQ, CXCR4,IL8,PF4V1, FPR1, CCL4involved in the chemotactic inflamm-atory pathwaychemokine and cytokine mediated; FOS,BCL2A1,CFLAR, MCL1involved inapoptosis signal pathway; PRKAR2B, GNAI3, STX11is involved in muscarinicacetylcholine receptors II and IV signaling pathway. The target gene has closelyrelationship with hsa-mir-182, hsa-mir-183and hsa-mir-25, hsa-mir-93, hsa-mir-363,hsa-mir-222and so on. Through drug treatment, finds it express different mRNA byclustering analysis, compared before with after treatment the gene is significantlyreduced, has transformed to the normal group trends.Conclusion:1Through RNA extraction combined with gene mRNA and MicroRNA chiptechnology, to find the different Gene and MicroRNA of Qi and Yin deficiencySyndrome, and establish the interaction network with the related gene, conduce toanalyse the internal molecular mechanism of Qi and yin deficiency syndrome of AIDS.2Through analyse the relation between the different MicroRNA and target gene of Qiand Yin deficiency syndrome group and healthy group, there are96target genes hasclosely relationship, the relationship between these genes and some MicroRNAclosely involved in response to trauma, regulation of cell proliferation, apoptosis, cellresponse to oxidative stress, etc.3After the Qi and Yin deficiency syndrome with HIV is treatment with Shengmaiyin, the different genes has transformed tends to normal group, this result suggest thatthese genes may be specificity for Qi and Yin deficiency of AIDS patients.This study analyzed the relationship of signaling networks between functional indifferences MicroRNA and their regulation target genes Qi and yin deficiencysyndrome in patients with HIV/AIDS. After the Chinese intervention outcomes gene,thus find related syndromes with AIDS or disease susceptibility related to thedevelopment specific genes, the next step binding studies proteomics, metabolomicsand other omics overall biological meaningful progress in nature.
