Objective:Copy the allergic rhinitis (Allergic rhinitis, AR) model in rats, observe the Shuyuwan decoction treatment behavior of AR rat index, serum total IgE and IL-4content in nasal mucosa, pathological morphology and aquaporin5(AQP5) expression, to explore the mechanism of treatment of AR, at the same time for the better applied to provide experimental evidence for clinical.Method:The SPF level of70SD rats, male, were randomly divided into:Normal control group, Model group, Xinqin granules group, Tongqiao biyan tablets group, High dose group, Middle dose group, Low dose group,10rats in each group. Except the Normal control group, each group with ovalbumin (OVA) to copy the model of AR rats. After the modeling, Model group and Normal control group rats were given oral administration of saline (1ml/0.lkg·d). High, Middle and Low dose group were given Shuyuwan decoction (the equivalent of crude drug32,16,8g/kg·d) by gavages. The positive control group were treated with Xinqin granule (5g/kg·d) and Tongqiao biyan tablets (1.8g/kg·d) by gavages. Each group was administered for14days. After the end of treatment, anesthetized rats’abdominal aorta blood extraction, using enzyme-linked immunosorbent assay (ELISA) detection of serum total IgE and IL-4content value. The rats were sacrificed, removal of nasal mucosa, for routine HE staining, detected the expression of protein AQP5in nasal mucosa with immunohistochemical SABC method.Result:After modeling, the integral of each behavior were higher than those in normal control group (P<0.01); the difference between the groups was not significant (P>0.05). After intragastric administration of14d, the treatment groups’behavior score were lower than the model group (P<0.01). Serum total IgE, IL-4content in the model group were higher than those in normal control group (P<0.01); the treatment group were lower than those in the model group (P<0.01); high, low dose group and Xinqin granules group, no significant difference (P>0.05); middle dose group than in Xinqin granule group decreased significantly (P<0.05); high, medium, low dose group and Tongqiao biyan tablet group, no significant difference (P>0.05). Under light microscope, the nasal mucosa of rats in the model group with the infiltration of inflammatory cells, edema, hyperemia, gland hyperplasia, part of epithelial tissue loss. In the treatment group compared with the model group, pathological morphology of nasal mucosa were significantly improved, in the medium dose group of tissue repair highlighted. In AQP5model group, the average optical density (MOD) was lower than that of normal control group (P0.05); in addition to Xinqin granules group, the treatment groups were higher than those in the model group (P<0.05); high, middle dose group was higher than that in tongqiaobiyan tablet group (P<0.01); low dose group compared with Tongqiao Biyan tablets group, the difference was not statistically significant (P>0.05). High, medium dose group was higher than that of low dose group (P<0.01); and the high, middle dose group comparison, no statistical significance (P>0.05).Conclusion:Shuyuwan decoction has significant therapeutic effect on AR rats. Decreasing the content of IL-4in serum, inhibit the synthesis of IgE, relieve inflammation, alleviate the inflammatory cells in nasal mucosa of infiltration, increase AQP5protein expression, regulation of nasal fluid metabolism, improve the sneeze, nasal itching, runny nose and other symptoms, may be one of the mechanisms of the good effects of the AR.
