Object:PIP and BBR, which are main component of piper longu L. and Coptis chinensisFranch. respectively, can reverse insulin resistance. To investigate the effects of PIPand BBR on the glucose metabolism disorder and underlying mechanism that mayregulated through AMPK, the cellular IR model of3T3-L1adipocytes wereestablished. This study may help to reveal the inherent scientific nature of Chineseherbal medicine and provide some evidence to clarify the mechanism of drugs whichcan improve IR.Method:Inducing IR cellular model of3T3-L1adipocyte by using1μM DEX afterdifferentiate into mature adipocytes of pre-adipocyte. Normal cell and IR model of3T3-L1were treated with PIP and BBR at different dose, AICAR and ROG were usedas positive control. The glucose intake of cells were determined after48h treatment ofdrugs, AMPK mRNA and protein were determined by RT-PCR and western blotting.The ratio ofAMP to ATP was investigated by HPLC.Results:1DEX-induced glucose consumption and glucose uptake were inhibitedsignificantly in3T3-L1adipocytes after incubation with DEX for48h, Whichdemonstrated the establishment of IR-adipocyte model.2Glucose consumption and uptake tests showed that in the presence of PIP orBBR, compared with IR group, glucose consumption and uptake rate displayed aremarkable increase in drug group.3Western blot assay showed that PIP and BBR markedly up-regulated the levelof P-AMPKα protein expression in IR-adipocytes. 4HPLC assay showed that PIP and BBR reduced the ratio of AMP/ATP afteraffecting IR-adipocytes for48h.Conclusion:IR-adipocyte model was established by adding1μM DEX for48hours. PIP andBBR could promote glucose metabolism and improve insulin resistance inIR-adipocytes. PIP and BBR might improve IR by targeting AMPK, increasing thelevel of AMPKα phosphorylation. Thus, this study, to a certain extent, reflects the roleof PIP in improving IR and the internal mechanism.
