Objective: Discussion Yishenyangyin mixture on renal function, renalpathological changes and the impact on P-STAT3expression, revealing itsmechanisms play a role in the efficacy and examining their safety, for the benefit ofYishenyangyin mixture treatment of lupus nephritis (LN) provides a theoretical basis.Methods: Two kinds of50different varieties of the same mice were randomly divided into5groups:normal group of C57BL/6J mices (0.9%saline).According to urinary protein content,MRL/lpr mices were randomly divided into model group(0.9%normal saline),traditional Chinese medicine group (Yishenyangyin mixture),chinese medicine combined hormone group (Yishenyangyin mixture+Prednisone acetate);and hormone group(Prednisone), each group of10mices,five perfusion drawn, five freshly drawn.The above groups continuous administration of four weeks.Detecting the expression of P-STAT3with Western blot method, observing the renal pathological changes with HE staining and immunofluorescence staining,BCA protein assay kit for urine protein, Detecting serum creatinine, urea nitrogen with colorimetric method.2.20C57BJ/6J mices were randomly divided into control group, Chinese medicine group, the maximum dose on mice by gavage for14days,observing the weight, detecting alanine aminotransferase, aspartate transaminase with enzyme linked immunosorbent assay, serum creatinine, and detecting blood creatinine and urea nitrogen with colorimetric method.Results:1.Urinary protein of MRL/lpr mices were significantly higher(p<0.05).In the Chinese medicine group, there was no significant difference on urinar y protein at1and2weeks (p>0.05), but urinary protein significantly reducedin the third and fourth weeks (p<0.05). Urinary protein decreased gradually at2,3,4weeks in Chinese medicine combined hormone group and hormone group、(p<0.05); compared with model group, urinary protein content between the treatment groups had no significant difference in the first week after administration(p>0.05).However, urinary protein content of each treatment group were significantly lower in the2,3,4weeks(p<0.05). Urinary protein was the lowest in Chinese medicine combined hormone group (p<0.05). Serum creatinine, blood ureanitrogen content in each treatment group were significantly lower (p<0.05), Chinese medicine combined hormone group decreased most significantly (p<0.05).After the administration of treatment, the expression of p-stat3of declining.Compared with the model group,the expression of p-stat3in traditional Chinese medicine group, hormone group decreased(p <0.05); reduce cases of Chinese medicine combined hormone group the most significant (p <0.05), while the expression of p-stat3in hormone group was higher than medicine group (p <0.05).HE staining showed that there are mild renal tissue partial mesangial cell proliferation in the three treatment groups, glomerular inflammatory cells decrease lgG immunofluorescence staining showed that there were lgG deposition in renal、tissue in the three treatment groups, but less in model group.2.The maximum volume of mice fed is120.34times bigger than clinicaladult (body weight is70kg). There was no significant difference between each group in body weight, liver and kidney function (p>0.05).Conclusion:1.Yishenyangyin mixture can improve the renal function and renal pathology of MRL/lpr mice, and the renal function improved obviously combined with hormone.2.Reduced expression of STAT3phosphorylation may be one of the targets forthe treatment of Yishenyangyin mixture of lupus nephritis.3.The acute toxicity on Yishenyangyin mixture mice had no obvious abnormalities on body weight, liver and kidney function, short-term use is safe, but it s till needs further study.
