ObjectiveThe study choose BMSCs, to exploring the Bushen Huoxue Decoction induc ed BMSCs to osteogenesis, providing theory for clinical use of traditiona1Chinese medicine to treat bone defect, delayed union or ununion, and Ion g immobilizlation disuse osteoporosis and other bone disease refractory, A nalyzing clinical data on intertrochanteric fractures patient, evaluation of the Bushen Huoxue Decoction in the treatment of Intertrochanteric fra ctures postoperation curative effect, providing theory for the treatment p ostoperation of intertrochanteric fractures.MethodExperiment:Serum containing Bushen Huoxue Decoction, then cultured in vitro to3rd generation. Then divided into5groups. Kidney medicine group, Promoting blood medicine group, All parties medicine group, Osteoinductive agent group, Blank group, after Induction the day of3th,6th,12th determining of ALP activity,15th measuring Ca2+concentration of Broth, measurin-g BMP R2proteinexpression after Western blot intervention72h, Observing the activity of osteoblasts.Clinical:60senile post-operation intertrochanteric fractures patient form The Third Affiliated Hospital of Guangzhou University of Traditional Chinese Medicine, then divided into2groups, comparative study methods of themselves were used, Bushen Huoxue Decoction group(group A) and Blank group(group B),30cases each group.Peripheral blood were taken at14d determining concentration ALP, Ca2+, observing post-operation hip joint Harris grade.Result 1. Using bone marrow adherent centrifugation to obtain higher High purity BMSCs;2. ALP concentration:at3th day, osteogenic group was statistically significant relative to the Promoting blood medicine group, Osteoinductive agent group and blank group(P<0.05);At6th day, no significant difference between the groups (P>0.05); At12th kidney group, all parties medicine group were statistically significant relative to the Promoting blood medicine group, Osteoinductive agent group and blank group (P<0.05)3. Ca2+concentration:at15th kidney group, All parties medicine group were statistically significant relative to the blank group(P<0.05).4. Bushen Huoxue Decoction treating postoperation of intertrochanteric fractures, Significantly increased serum ALP concentrations, There are sta tistically significant between the two groups(P<0.05);and No significant ly increased Ca2+concentration, There was no statistically significant bet ween the two groups (P>0.05)Conclusion1. In vitro whole bone marrow adherent cell separation can obtain high purity of BMSCs.2. Bushen Huoxue Decoction eontaining serum can obviously Promote their osteogenesis diferentiation of BMSCs.3. Bushen huoxue Decoction can relieve postoperation of intertrochanteric pain, Promote the recovery of limb function and fracture healing.4. Bushen huoxue Decoction for intertrochanteric fracture medical expense is low, the curative effect is exact and reliable.
