Objective:To observe the effect of Inonotus obliquus on the level of blood glucose and oxidative stress in patients with type2diabetes, and investigate the relationship of blood glucose control level and oxidative stress indicator nitrotyrosine with the expression of HO-1protein.Methods:The experimental subjects were divided into three groups, namely the normal control group, the type2diabetes group receiving simple insulin therapy and the type2diabetes group receiving insulin as well as Inonotus obliquus powder daily, with30cases in each group. After4months, venous blood was collected to observe whether there was statistical difference among three groups in biochemical indicators such as cholesterol, triglyceride, fasting blood glucose and HbAlc, in serum nitrotyrosine level and in the expression of HO-1protein and HO-1mRNA in peripheral blood mononuclear cells (PBMCs). The serum nitrotyrosine level of all the subjects was detected by ELISA. The expression of HO-1mRNA in PBMCs was detected by RT-PCR. The expression of HO-1protein in PBMCs was detected by western blot. As for the statistical methods, LSD-t was used for the comparison between any two groups, ANOVA was used for the comparison among three groups and chi-square test was used for the comparison of enumeration data. Pearson’s linear correlation was adopted for the correlation analysis and partial correlation analysis was performed with variables controlled.Results:The level of nitrotyrosine showed difference (P<0.05) and a gradual increase in the normal control group, Inonotus obliquus group and simple insulin group. With the effects of blood lipid removed, the level of nitrotyrosine showed positive correlation with that of blood glucose. The level of HO-1protein and HO-1mRNA expression showed difference (P<0.05) and a gradual increase in the normal control group, Inonotus obliquus group and simple insulin group. In the correlation analysis, the nitrotyrosine of Inonotus obliquus group and simple insulin group showed positive correlation with blood glucose, HbAlc and blood lipid (P<0.05), and showed significant positive correlation with HO-1protein and HO-1mRNA (P<0.01). The HO-1protein showed positive correlation with blood glucose, HbAlc and blood lipid (P<0.05), and showed significant positive correlation with HO-1mRNA (P<0.01). In the partial correlation analysis, with the effects of blood lipid removed, the nitrotyrosine of Inonotus obliquus group and simple insulin group showed positive correlation with blood glucose and HbAlc (P<0.05), and showed significant positive correlation with HO-1protein and H0-1mRNA (P<0.01). With the effects of blood lipid, blood glucose and HbAlc removed, the expression level of HO-1protein can not show correlation with nitrotyrosine level.Conclusion:The level of oxidative stress and the level of blood glucose were gradually increased in the normal control group, Inonotus obliquus group and simple insulin group, suggesting that Inonotus obliquus may have good in vivo antioxidant effect and glucose-lowering effect in patients with type2diabetes. Endogenous antioxidant enzyme H0-1protein was induced in the Inonotus obliquus group and simple insulin group, and the level of induced HO-1was positively correlated with the level of oxidative stress, suggesting that HO-1protein may play a very important role in the oxidative stress of patients with type2diabetes.
