Purpose:1.To observe the different doses of Russian Boschniakia Herb extracts which influence the level of TNF-a,IL-10of ARDS rat serum.2.To research the Russian Boschniakia Herb extracts of phosphorylated p38MAPK in ARDS rat lung tissue (p-p38MAPK) protein expression.3.To explore the protection effect of Russian Boschniakia Herb extracts of ARDSMethod:Divide SD male rats which average weight is50(200±20) g, adaptability raised after a week into5groups, each group of10, the blank group, model group, low dose Russian Boschniakia Herb group, medium dose Russian Boschniakia Herb group, high dose Russian Boschniakia Herb group. At first6days intragastric1ml/100g saline to blank group and model group rats per day, meanwhile, intragastric125mg/kg,250mg/kg and500mg/kg dose of Russian Boschniakia Herb extracts to low, medium and high dose Russian Boschniakia Herb. In the sixth day, peritoneal postanesthetic all animal with urethane (1.0g/kg), give blank group5ml/kg of saline by tail intravenous. in the model group,low-dose Russian Boschniakia Herb group, medium-dose Russian Boschniakia Herb group, high-dose Russian Boschniakia Herb group inject oleic acid0.1ml/kg by tail intravenous. After three hours, observe the general condition of rats,and execute them. Calculated lung wet/dry weight ratio (W/D) of left lung; observe the right lung tissue pathological changes with HE staining; detect the serum concentration of TNF alpha and IL-10with ELISA method; detect the protein expression of p-p38MAPK of right lung tissues by western blot method.Consequences:1.Compared with the blank group, model group rats left lung W/D increased significantly (P<0.05); Compared with model group, Russian Boschniakia Herb, high dose group of W/D of the left lung of rats significantly (P<0.05).2.The result shows:the histopathologic HE staining, the blank group rats lung tissue structure is clear, thin bronchial mucosal epithelial integrity, false stratified columnar epithelium cells arranged orderly, alveolar cavities is clear, interval complete without edema, no obvious inflammatory exudation:Model group rats lung tissue structure is incomplete, and alveolar walls, alveolar cavities seen predominantly neutrophils infiltrating inflammatory cells, and a large number of exudative edema fluid, protein pulmonary interstitial capillary expansion, congestion, alveolar interval obvious thickening and edema; Russian Boschniakia Herb each dose group of rat lung tissue structure basic integrity and alveolar cavity edema, neutrophil infiltration, and thickening of alveolar interval compared with model group. Russian Boschniakia Herb lower inflammatory lesions in high dose group, middle dose group of light.3.Compared with the blank group, the concentration of TNF-a, IL-10of model group rats has been increased significantly (P<0.05); Compared with model group, the concentration of TNF-a, IL-10of medium and high dose Russian Boschniakia Herb group significantly decreased (P<0.05).4.The expression of P-p38MAPK protein in lung tissue of blank group rats is very weak, the expression of p-p38MAPK protein in model group increased significantly, compared with the model group, the expression of p-p38MAPK of each dose Russian Boschniakia Herb group decreased obviously. Conclusions:When the ARDS occurred, Russian Boschniakia Herb extract has obvious protective effect on normal lung tissue.1.Russian Boschniakia Herb extract,inhibit the expression of serum TNF-aand IL-10;2.Russian Boschniakia Herb extracts inhibit the expression of p-p38MAPK in lung tissues, the reason may be associated with the regulation of Russian Boschniakia Herb on the MAPK pathway;3.If the dose of extracts of Russian Boschniakia Herb is125~500mg/kg, when the ARDS occurred the protective effect in normal lung tissue is proportional to the dose.
