Background: Bullous pemphigoid (BP) is the most common subepidermal autoimmuneblistering disease, characterized by tense blisters and erosions on skin or mucous membranes.Stroke is a sudden neurological deficit due to localized brain ischemia or hemorrhage.Previous case reports and population-based epidemiological studies have suggested arelationship between BP and neurological diseases, among whom, stroke is one of the mostrelevant comorbidities. However the researches about the genome-wide association study(GWAS) of BP remains zero so far. We therefore hypothesized there might be sharedsusceptibility loci between BP and stroke.Objective:1) to evaluate the prevalence of stoke in Chinese BP patients;2) to investigatethe common pathogenesis between them.Methods:1)160BP cases were recruited to evaluate the prevalence of stroke in ChineseBP patients;2)74single-nucleotide polymorphisms (SNPs) within41loci which showedgenome-wide significant associations with stroke in Caucasian population were identified. Weexcluded those SNPs whose minor allele frequency in Chinese population are0and SNPswhich are located in major histocompatibility complex region, ultimately48SNPs associatedwith stroke in Caucasian population were successfully designed and genotyped in the160BPpatients and564controls using Sequenom MassArray systemResults:1) The prevalence of BP cases with stroke was15%(24cases), which was muchhigher than4.1%in Chinese people aged65-74years;2) Logistic regression was performed to test the genotype association in each SNP using Plink v1.07.14. After adjusting for age andgender, all48SNPs showed no significant association with susceptibility to24cases withstroke, as well as to136cases without stroke compared with564healthy controls in ChinesePopulation (p value>0.05).Conclusion: We confirmed the relationships between stroke and BP in Chinese population,but no remarkable association with susceptibility SNPs of stroke to BP cases in Chinesepopulation.
