Objectives:In recent decades, along with the developing of the basic and clinical researchs on breast cancer, the pathogenesis and treatment of breast cancer have made great progress. However the factors of influencing breast cancer treatment and prognosis and the relationship of these factors are still not clearly, especially the triple-negative breast cancer (TNBC) as a special kind of breast cancer which has high malignant rate and poor prognosis, so far, there is no effective drugs for it, since it lacked the effective drug targets. The aim of this study was to detect the the expression of BAG-1, EGFR and PARP-lin breast cancer and analysis their expression levels in breast cancer and peritumoral tissues, and to explore the the relationship between the their expression level and clinical and pathological indicators, and further to investigate the significance of BAG-1, EGFR and PARP-1expression status in the diagnosis, treatment and prognosis of breast cancer.Methods:breast tissue microarray were purchased from Shanghai Outdo Biotech Co., itd. Each breast tissue microarray contained119cases of breast cancer specimens and40cases of peritumoral specimens. The expression of BAG-1, EGFR and PARP-1in tissue microarray of breast cancer and peritumoral tissues were detected by immunohistochemical staining, and explore the relationship between the their expression level and clinical and pathological indicators. And further to analysis the correlation of the status of BAG-1, EGFR and PARP-1and prognosis of breast cancer.Results:1. The positive expression rate of BAG-1, EGFR and PARP1protein in119cases of breast cancer tissue samples was95.80%(114/119),31.09%(37/119) and57.14%(68/119), respectively. And the positive expression rate of BAG-1, EGFR and PARP-1protein in40cases peritumoral specimens was73.53%(25/34),13.51%(5/37) and22.86%(8/35),respectively(individual peritumoral specimens taken off).2. The positive expression of BAG-1protein was correlation with tumor size, histological grade, the expression of ER, PR, HER2(P<0.05); the positive expression of EGFR protein was correlation with tumor size, the expression of ER, PR (P<0.05); the positive expression of PARP-1protein was correlation with histological grade, number of lymph node metastasis, ER expression (P<0.05).3. There were significance differences of the expression of BAG-1, EGFR and PARP-1in different breast cancer subtypes. The expression of BAG-1in HR(+)/HER-2(-) is more than other subtypes, and the expression of EGFR and PARP-1in TNBC is more than other subtypes, respectively(P<0.05). Spearman correlation analysis showed that BAG-1expression and PARP-1expression were positively correlated in TNBC (r=0.651, P=0.001).4. Univariate analysis of prognostic factors in breast cancer patients show that tumor size, histological grade, number of lymph node metastasis, the expression of EGFR and PARP-1, the subtypes are the relevant factors(P<0.05).5. Multivariate analysis results showed that histological grade, number of lymph node metastasis, the expression of EGFR and PARP-1, the subtypes may be as an independent prognostic factor of breast cancer(P<0.05).Conclusions:1. The positive expression rate of BAG-1, EGFR and PARP1protein in breast cancer tissue samples than that in peritumoral tissue. The positive expression of BAG-1protein is correlation with tumor size, histological grade, the expression of ER, PR, HER2; EGFR is correlation with tumor size, the expression of ER, PR; and PARP-1was correlation with histological grade, number of lymph node metastasis, ER expression.2. There were significance differences of the expression of BAG-1, EGFR and PARP-1in different breast cancer subtypes. The results of BAG-1expression and PARP-1expression were positively correlated in TNBC showed that BAG-land PARP-1may be as the targets of TNBC.3. BAG-1and PARP-1proteins may be as an independent prognostic factor of breast cancer. The high expression status of BAG-1, EGFR and PARP-1may be as the biomarkers of diagnosis, treatment and prognosis of breast cancer.
