Liver disease is one of the most common diseases affecting human health, various harmful liver injury causes, its are clinical expression is:necrosis of liver, fatty liver, cholestasis, liver fibrosis, cirrhosis and liver cancer. According to the syndrome differentiation and treatment of common pathogenesis of liver injury, a common therapeutic drugs, traditional Chinese medicine, and in vivo animal models of liver injury, from making method of somatic liver injury model of experimental research in this paper;ObjectiveThrough in vitro liver cell injury model, the medicated serum intervention armored decoction, to investigate the protective effect of Cassia mimosoides Decoction on liver cells, for cassia mimosoides drugs in the prevention and treatment of liver damage disease provide experimental basis.Methods1、Culture of L02liver cells, using a final concentration of hydrogen peroxide solution of0.6mmol/L establishment of liver cell injury model, cells were divided into6groups:normal control group, model control group, positive control group, low, medium, high concentration group, in addition to the normal group, each group of rats1.5hours after the termination model, each adding serum the corresponding1.5hours after termination of the intervention, the content of hepatic cell homogenate enzyme mark instrument to detect ALT, AST, GSH, MDA, GSH-px content detection by UV spectrophotometry, the cell MTT method to measure the cell viability, and cell morphology were observed in liver;2、Culture of L02liver cells, with the final concentration of carbon tetrachloride solution of8mmol/L establishment of liver cell injury model, cells were divided into6groups:normal control group, model control group, positive control group, low, middle, high dose group, in addition to the normal group, each group of rats6hours after termination of molding, the group added serum role of the corresponding1.5hours after termination of the intervention, the content of hepatic cell homogenate enzyme mark instrument to detect ALT, AST, GSH, MDA, GSH-px content detection by UV spectrophotometry, the survival rate of the cells was measured by MTT method and cell, liver cell morphology was observed.ResultsMTT results showed that:the high dose group of cell survival rate was significantly increased, compared with the model group, there was significant difference, low, middle dose group compared with the model group, no statistical significance, relatively high concentration group and positive control group, the difference was not significant; the determination of ALT, AST display:given the drug containing serum intervention, in high dose group, ALT, AST decreased significantly, compared with the model group, there was significant difference. Low dose group compared with the model group, no statistical significance; MDA showed that, given serum intervention, the content of MDA in high dose group were significantly decreased, compared with the model group, there is statistical significance, the low, middle dose group compared with the model group, high dose group was not statistically significant, compared with the positive control group, the difference was not significant; the determination of GSH, GSHpx showed:serum containing Kaixin Capsule high dose group, content of GSH, GSHpx increased significantly, compared with the model group, there is statistical significance, the low, middle dose group compared with the model group, no statistical significance, the high dose group compared with the positive control group, the difference not significant. Liver cell morphological observation showed:the normal group, the more number of liver cells with smooth surface, transparent degree is strong, the cell membrane integrity, clear edge, clear, and the model group liver cell surface folds, the membrane is not complete, transparency, darker than the normal group, the quantity is little, the edge is not clear, low profile fuzzy; the middle dose group, surface smoothness is poor cell, fewer, transparency, dim, blurred edges, outline is not clear; the high dose group of cell surface smooth, edge, contour visible, bright bright number compared with the model group, compared with normal group; positive control group, cell surface smooth, edge, contour visible, transparent with the model group compared with normal group and less number of bright. The serum iron Decoction high dose can improve the morphology and structure of liver cells.ConelusionEstablishment of a liver cell model in vitro, determined the serum iron decoction has a protective effect on liver cells, improve the survival rate of liver cells, reduce the liver cells to release ALT, AST, increase the content of intracellular GSH, GSHpx, decrease the content of MDA, which may have scavenging antioxidant effects of free base, associated with serum containing iron decoction, pending further study and discuss deeply.
