OBJECTIVETo detect and investigate the effects of endogenous hydrogen sulfide on theexpression of angiotensin II type1in rat with hepatic fibrosis induced by carbontetrachloride.METHODSFifty-six Wistar rats were randomized into four groups: nomal control group,model group, NaHS group, PAG group. Except the rats in nomal group,hepatic fibrosiswas induced using carbon tetrachloride(CCl4) in rats. For rats in PAGgroup,DL-propargylglycine, an inhibitor of cystathionine-β-synthase, was intraperitoneallyinjected. For rats in NaHS group, sodium hydrosulfide, an H2S honor, was intraperitoneallyinjected. An equal volume of normal saline was injected in the control and model groups.All rats were killed at week3or week4after modeling. The level of serumhyaluronidase(HA), laminin protein(LN), III procollagen(PcIII),IV collagen(cIV) weredetected by ELISA. The level of serum ALT, AST, albumin(ALB value) were detectedby. The liver mRNA expression level of CSE was detected by real-time fluorescencequantitative RT-PCR. The liver expression of AT1was detected by western blotting, atthe same time plasma H2S was measured.RESULTS1)The same point in time, histological damage scores in model group, NaHS groupand PAG group was significantly higher than normal control group. Histological damagescores in PAG group was significantly higher than model group. NaHS group wassignificantly lower than model group(P<0.05).2) The level of serum HA, LN, PcIII, cIV, ALT, AST in model group, NaHS roup and PAG group were significantly higher than normal control group. The level of serum HA,LN, PcIII, cIV, ALT, AST in PAG group were significantly higher than model group. NaHSgroup were significantly lower than model group,the change of ALB is oppsite(P<0.05).3) The liver mRNA expression of CSE in model group, NaHS group and PAG groupwas significantly higher than normal control group. The liver mRNA expression of CSE inPAG group was significantly higher than model group. NaHS group were significantlylower than model group(P<0.05).4)The liver tissue expression of AT1in model group, NaHS group and PAG groupwas significantly higher than normal control group. The liver tissue expression of AT1inPAG group were significantly higher than model group. NaHS group was significantlylower than model group(P<0.05).5) The plasma H2S in model group, NaHS group and PAG group was significantlyhigher than normal control group. The plasma H2S in PAG group were significantly higherthan model group. NaHS group was significantly lower than model group(P<0.05).CONCLUTIONEndogenous H2S was closely related with CCl4induced hepatic fibrosis in rat, and has aanti-fibrotic effect within a certain range. Moreover, H2S reduced the liver tissueexpression of AT1, which may associate with the delayed progress in hepatic fibrosis.
