Objective:Study of correlation between serum amyloid A1level (serum amyloid a, SAA1) and the rs12218gene polymorphism with type2diabetes mellitus(T2DM) and nephropathy (DN) relevance.Methods:Select of275control cases from Yanbian area,Include normal control groups (normal glucose tolerant, NGT)50cases and T2DM piatients225case. According to urinary albumin excretion rate (UAER), T2DM group divid into Diabetic Kidney disease-free group (NDN)70cases and diabetic nephropathy (DN)cases140. Use the method of (ELISA) Determination of serum levels of SAA sample and use the Fluorescent sequencing method Determination of gene sequence, sample DNA ampliation、 purification and DNA sequencing. Genotyping by sanger method.. Ria method to determine the concentration of urine albumin, then use SPSS17.0statistical software statistical analysis of data.Measurement data is owed by mean±standard deviation (x±s), one-way analysis of variance is used for comparison among groups. The genotype and allele frequency are compared by chi-square test of the relationships between genotype and allele frequency. Logistic regression to analysis SAA and Risk factors for DN disease screening.Results:1.In Yanbian area, the level of SAA1in Type2diabetes mellitus is higher than normal control groups, and the level of SAA1in DN group is higher than NDN group, significant difference in the distributions of two groups (P<0.05).2.The level of SBP, DBP, FPG,2hPG, HbAlc, CREA in Type2diabetes are higher than the control group, there are significant differences between them(P<0.05).3.There are positive correlation between the level of SAA1and the SBP, DBP, FPG,2hPG, HbAlc, CREA in diabetic nephropathy, has Statistical significance(P<0.05).4.1n YanBian area, Korean and Han population, SAA1gene rs12218allele gene frequency compare with Hap Map database (SS40072010) of Europeans (CEU)、 China Han in Beijing (HCB),、 Tokyo people (JPT) and Africa Yoruba (YRI), rs12218(A) has no national difference (p>0.05), and Korean rs12218A allele gene frequency higher thanCEU (p<0.01), but in HCB^JPT has no significiant difference (p>0.05)。5. rs12218-G allele gene frequency in NDN group higher than NGT group (32.3%>16.0%, P=0.045),and DN group significiant higher than NDN group(33.2%>32.3%, P=0.046)。The dominant mode (AG+GG:AA), has AG+GGgene NDN risk was1.673times higher than NGT group(P=0.04, OR=1.673,95%CI:0.76-3.69), and has AG+GG gene get DN risk was1.46times higher than NGT group(P=0.046, OR=1.673,95%CI:0.67-3.35)。6.The levels of SAA1with AG+GG gene in NGT、NDN、DN group is higher than with AA gene, has significant difference between them (P<0.05).7.The polymorphisms of NGT group、NDN group、DN group, there were no significiant differences (P>0.05), in dominant mode there has no significiant difference (P>0.05),recessive mode also has no significiant differences (P>0.05)。8.PBG、SAA1level and AG+GG gene are the risk factors of DN.Conclusion:1. There has positive correlation between SAA1level and T2DM、DN.2. SAA1gene rs12218-G allele susceptibility gene with DN.3. SAA1gene rs12218polymorphism has national difference.
