ObjectiveIn this study, non-small cell lung cancer (NSCLC) and adjacent normal lung tissue is used by the method of immunohistochemistry to investigate the expression of cell division cycle42(Cdc42) and WASP family verprolin-homologous protein1(WAVE1) and their clinical significance.MethodsSpecimens were selected from Tianjin Chest Hospital during January to December in2008, resected NSCLC specimens (106cases) and adjacent normal lung (46cases) fixed in10%formalin and embedded in paraffin. The method of immunohistochemistry is used to investigate the expression of Cdc42and WAVE1. Differences of expression of Cdc42and WAVE1in NSCLC tissues and normal lung tissues were analysed; correlations of expression of Racl and NF-κB p65to clinical pathological parameters and prognosis and their correlations were analysed using SPSS17.0.Results(1) Cdc42and WAVE1both expressed in NSCLC tissues and normal lung tissues. Cdc42expressed mainly in the cytoplasm, and the expression intensities of Cdc42were85.85%(91/106) and23.91%(11/46). WAVE1expressed mainly in the cytoplasm, and the expression intensities were82.08%(87/106) and17.39%(8/46). Both Cdc42and WAVE1expression in NSCLC tissues was significantly higher than in normal tissues, and the expression in normal tissues was low.(2) Univariate analysis of variance found that the expression of Cdc42in NSCLC tissues was correlated significantly with differentiation, TNM stage and lymph node metastasis(P<0.05). The expression of WAVE1in NSCLC was correlated significantly with TNM stage and lymph node metastasis(P<0.05or P<0.01).(3) Spearman’s rank correlation found that the expression of Cdc42and WAVE1in NSCLC demonstrated significantly positive relation (r=0.469, P<0.01)(4) The3-year survival rates with Cdc42high expression(44.16%) were lower in patients than those with low expression(72.41%), and the3-year survival rates were lower in patients with WAVE1high expression(39.44%) than those with low expression(77.14%), and both of them were considered significant (P<0.01). Expression of both Cdc42and WAVE1, either Cdc42and WAVE1, the low expression of both Cdc42and WAVE1were38.71%,41.67%and85.00%, also considered significant(P<0.01).According COX multivariate regression analysis, lymph node metastasis, Cdc42and WAVE1common high expression were independent factors affecting patient prognosis with NSCLC, and gender, age, histological type, tumor differentiation and tumor size were with no significant relationship.ConclusionIn NSCLC tissues, the expression of Cdc42and WAVE1showed a good correlation. They may act in concert and play an important role in NSCLC. Detecting the expression of Cdc42combined with WAVE1in NSCLC tissue may play a suggestive function of clinicopathological features and prognosis.
