BackgroundWorldwide, there are tens of thousands of patients with liver fibrosis, with the progression of these patients may develop cirrhosis, liver cancer, liver failure and even eventually die, although the study of liver fibrosis has been made great progress, but because of complex mechanisms of hepatic fibrosis, still no more effective treatment. In recent years, with the development of molecular biology and gene therapy technology, more and more scholars recognize that liver fibrosis is a result of a variety of cytokines, and these cytokines constitute a complex network system acts on the liver cells, leading to liver cell damage. This preliminary study showed that subjects American cockroach extract has anti-experimental liver fibrosis, the role of this effect in a rat model of how the mechanism, we need further study.objectiveCarbon tetrachloride-induced liver fibrosis in rats, further observation American cockroach extract (APA) on rat liver fibrosis results, preliminary study American cockroach extract on hepatic fibrosis in ratsMethods70male SD rats were randomly divided into seven groups, namely the control group, model group, APA high-dose group, APA dose group, APA low-dose group, Panax salvia and colchicine positive control group. In addition to the control group, the other groups of rats were injected in the back of the neck20%carbon tetrachloride olive oil solution,0.05ml/10g,2times/week, for seven consecutive w. Meanwhile in modeling, blank control group and model group to distilled water daily; APA each dose group, Panax SM group and colchicine group fed the appropriate drugs were tested, seven consecutive w. The rats were sacrificed after the last administration, routine testing of liver function; liver tissue specimens, the line hematoxylin-eosin (HE), Masson staining; Immunohistochemical detection of liver tissue by transforming growth factor-β1(transforming growth factor-β1, TGF-β1), tissue inhibitor of metalloproteinase-1(tissue inhibitor of metalloproteinase-1, TIMP-1), a-smooth muscle actin (a-smooth muscle actin, a-SMA), the nuclear transcription factor-KB (nuclear transcription factors-KB, NF-κB); radioimmunoassay hyaluronate (Hyaluronic acid, HA), laminin (Laminin, LN), Ⅲ procollagen (Procollagen-Ⅲ, P-C Ⅲ), IV collagen (Type IV collagen, C IV). Experimental data were analyzed using statistical software SPSS17.0.Results1American cockroach extract can improve the mental state of hepatic fibrosis in rats, activity, reaction to the outside world, fur color, etc. In general, where American cockroach extract more significant improvement in the high-dose group;2treatment groups in rat liver specimens naked eye, color, surface smoothness, edge, texture, swelling, etc. are better than the untreated group, which American cockroach extract high-dose group, better than the other treatment groups;3American cockroach extract can reduce liver fibrosis serum alanine aminotransferase (alanine aminotransferase, ALT), aspartate aminotransferase (aspartate aminotransferase, AST) and other indicators of liver function, may also reduce liver fibrosis in rats serum HA, LN, P-C Ⅲ, C IV four indicators of liver fibrosis, especially in the high-dose group was significantly lower.4American cockroach extract can improve liver fibrosis damage, reduce G, S pathological score, but the difference between the treatment groups was not statistically significant.5American cockroach extract can reduce liver fibrosis in rat liver tissue TGF-β1, TIMP-1a-SMA expression,, NF-κB, where American cockroach extract high doses significantly lower.Conclusion1.various therapeutic drugs on the mental state of hepatic fibrosis in rats, activity, reaction to the outside world, fur color, etc. all have to improve the role of the general situation, in which high doses of American cockroach extract improved more significantly;2.American cockroach extract on hepatic fibrosis in rats liver damage, liver damage has a protective effect; including high doses of American cockroach extract more apparent protective effect; 3. American cockroach extract can reduce liver fibrosis serum HA, LN, P-C Ⅲ, C Ⅳ four indicators of liver fibrosis, particularly the role of American cockroach extract high doses is more obvious. And improve liver fibrosis damage, reduce G, S pathological score, indicating American cockroach extract has therapeutic effects in rat liver fibrosis.4. American cockroach extract on experimental hepatic fibrosis may be related to lower levels of TGF-β1expression, leading to decreased expression of TIMP-1gene, resulting in decreased synthesis of ECM, delaying the progression of liver fibrosis.5. American cockroach extract antifibrotic mechanism of action by inhibiting IκB kinase activation, thereby inhibiting NF-κB activation and the shift to the nucleus, preventing the activation of hepatic stellate cells, inhibition of TNF-a, IL-6, intercellular adhesion molecule (ICAM)-1, expression of transforming growth factor (TGF)-β1, such as NF-κB downstream of proinflammatory cytokines, thereby inhibiting cytokine cascade between each amplification effect, and ultimately prevent the development of hepatic fibrosis...
