Objective:Investigate of RS1800247and RS1543297locus single nucleotide polymorphisms (SNPs) correlation with type2diabetes glucose, lipid, bone metabolism.Methods:According to their bone mineral density level,217patients with type2diabetes were divided into two groups:normal bone mineral density group and low bone mineral density group. By PCR-RFLP analysis techniques detection of RS1800247and RS1543297locus gene polymorphism.Results:(1) The frequencies of genotypes in217type2diabetic patients were as follows:TT(59.4%), CT (33.6%),CC (6.9%) for RS1800247polymorphism; TT (30.4%), CT(49.3%), CC (20.3%) for RS1543297polymorphism; The genotype distribution of each polymorphism site was met with Hardy-Weinberg equilibrium (P>0.05).(2) There were no significant differences in the genotype and allele distributions of RS1800247and RS1543297polymorphism (P>0.05).(3) RS1800247:compared with TT and CT+CC genotypes between the two groups, homeostasis model assessment insulin secretion index (Homa-β) had significant difference (P<0.05).(4) RS1543297:Compared with TT and CT+CC genotypes between the two groups, insulin sensitivity index (ISI), homeostasis model assessment of insulin resistance index (Home-IR) had significant difference (P<0.05).Conclusions:The promoter region of OC gene RS1800247sites genotype differences of race and region. RS1800247sites may be associated with type2diabetes glucose metabolism indicators, Among patients carrying the T allele may have effects on islet beta cell failure, patients carrying the C allele may be a risk factor for pancreatic beta cell function decrease. RS1543297sites may be associated with type2diabetes glucose metabolism indicators, patients carrying the T allele may have strong sensitivity insulin, Patients carrying the C allele may have insulin resistance. The study did not find RS1800247, RS1543297loci associated with type2diabetes, bone metabolism and lipid metabolism.
