For the determination of quercetin on apoptosis of human gastric cancer cell lineSGC-7901cells to produce the effect of different concentrations of quercetin (20,40,80μmol/L) pretreated SGC-7901cells, the Annexin V/PI staining, the cell apoptosis rate weredetected by flow cytometry.4. For quercetin determination effect of quercetin on the expression of Cav-1proteinwas treated by different concentration (20,40,80μ mol/L) pretreated SGC-7901cells, theexpression changes of Cav-1cells were detected by Western blotting method.Results1. Quercetin can inhibit the proliferation of gastric cancer SGC7901cells: theapplication of MTT method to observe and analyze the quercetin effect on survival rate ofgastric cancer SGC7901cells, TG and RAW264.7cells respectively by5,10,20,40,80μ mol/L three concentration of common culture, in24,48h,72h three time points to measurethe cell survival rate, results show that, in each period is20,40,80umol/lQue on gastriccancer cell SGC7901inhibited compared with the control group had significant difference(P<0.05), in a dose-dependent manner.2.Quercetin can influence SGC-7901gastric cancer cells: morphology was observedby inverted microscope number in the control group with normal gastric cancer SGC-7901cells were cuboidal cells, shape, spacing, cells were monolayer adherent growth, growthstretch, shedding almost invisible; the culture medium after treatment with quercetin, cellvolume became small and round, crinkle, shedding, visible cells suspended in culturemedium, the nuclear color deepens, refraction enhanced, cell membrane integrity butfoaming phenomenon, it indicated that the cell morphological changes of apoptosisappeared. Compared with the control group, with different concentrations of quercetintreatment, cells were increased with increasing concentration of the apoptosis degreebecome more and more serious, show morphological changes and cell apoptosis ofquercetin concentrations also showed dose-dependent.3. Quercetin can promote SGC-7901cell apoptosis in gastric carcinoma: Quercetinpretreatment of cells, stained with Annexin V-FITC/PI double staining, detected theapoptosis rate of SGC-7901cells in gastric cancer by flow cytometry under. The resultsshowed, in control group the apoptosis rate of (5.62±1.65)%; after Que (10,20,40and80umol/l), total apoptosis rate was (11.39±1.13)%,(32.69±1.40)%,(66.72±1.96)%,(72.39±1.30)%, compared with compared with the control group increased significantly(P<0.05), in a dose-dependent manner.4. Quercetin can inhibit the expression of Cav-1protein and SGC-7901in gastriccancer cells: SGC-7901cells were treated with quercetin treatment with differentconcentrations of24h, we use the expression changes of caveolin-1cells were detected inwesternblot. The results showed that the expression of caveolin-1protein in experimentalgroup, each group were significantly lower than the control group, the content of relativeprotein10,20,40,80umol/lQue treatment group were decreased by46.2%,62.7%,95%and98.7%compared with the control group (P<0.05), the difference was statisticallysignificant. The results suggest that quercetin can inhibit the expression of caveolin-inhuman gastric cancer cell line SGC-79011protein.Conclusion1. Quercetin can inhibit the proliferation of SGC-7901and influence cell morphology.2.Quercetin can promote SGC-7901cell apoptosis in gastric carcinoma. 3. Regulating effects and mechanism of quercetin on gastric cancer SGC-7901cellsmay be associated with reduced caveolin-1expression.
