Objective We investigate the expression of Toll like receptor6protein in peripheral blood mononuelear cell in children with Henoch-Schonlein purpura(HSP),and its correlation with Th1、Th2and Th17cell, to explore the relationship between Toll-like receptor6and the pathogenesis of HSP.Methods Totally42children with Henoch-Schonlein purpura were enrolled in the studyAnother30healthy age-matched children as controls.The relative expression level of MyD88mRNA in peripheral blood mononuclear cells was detected by real-time fluorescent PCR.Protein levels of Toll-like receptor6was detected by flow cytometric analysis.Serum levels of IFN-y-. IL-4and IL-17in plasma were determined by ELISA.Results1.The levels of Toll-like receptor6proein and MyD88mRNA of the HSP children were significantly higher than the control group (P<0.01)2.The levels of IL-4、IFN-y and IL-17in plasma of HSP children were significantly higher than the controls,(t’=3.44, P<0.01; t’=2.44, P<0.05; t’=2.64, P<0.01),and the ratio of IFN-y/IL-4in plasma of HSP children was lower than the control group(t’=2.27, P<0.05).3. The mRNA expression of MyD88showed a positive correlation with expression of Toll-like receptor6protein of CD4+T in HSP children (r=0.79; P<0.01); Toll-like receptor6had positively correlated with IL-4(r=0.69, P<0.01); and negatively correlated with the ratio of IFN-γ/IL-4ratio(r=-0.38, P<0.05); and positively associated with IL-17(r=0.36, P<0.05); had no correlation with plasma IFN-y(P>0.05)Conclusions1.The expression levels of TLR6protein and MyD88mRNA in PBMCs were remarkably increased with the HSP children.2.The Thl/Th2imbalance exists in HSP children, and the Th2relatively prevails over Thl. Th2functions increased correlates closely with high expression of TLR6on PBMCs.3.There is express of Th17and had positive correlation with the TLR6protein4.The activation of TLR6protein may mediate immune pathogenesis of HSP by up-regulation Th2and Th17immune responses.
