Objective: This study detected the expression of TopoIIα in benign and malignant breastdiseases,analysed the relationship between the characteristics of the TopoIIα expressionand the clinical and pathologic features in breast cancer tissues. To investigate the role ofTopoIIα expressing in the mechanism of occurrence, development and metastasis withbreast cancer, and provide theoretical basis for the evaluation, therapy and prognosis ofbreast carcinoma.Methods: The expression of TopoIIα was detected using immunohistochemistry in71infiltrating ductal carcinoma tissues,23intraductal carcinoma tissues,20benignhyperplasia of epithelium tissues and30mastopathia normal tissues.To make an analysison the expression of TopoIIα in infiltrating ductal carcinoma with patients’ clinicalfeatures such as age, tumor size, clinical stage, histological grade, lymph node metastasis,and pathologic features such as the expression of ER, PR, HER-2and Ki-67.Results: The positive expression of TopoIIα was found20.0%of mastopathia normaltissues,25.0%of benign hyperplasia of epithelium tissues,43.5%of intraductal carcinomatissues,62.0%of infiltrating ductal carcinoma tissues. Topo Ⅱα has a higher positiveexpression rate(57.4%) in breast cancer tissues than those in normal tisses of mammarygland(22%),(P<0.01). Statistically significant differences and positive correlations inTopo Ⅱα expressionin infiltrating ductal carcinoma tissues between the ones with lymphnode metastases(72.5%) or not(48.3%),(P<0.05). Statistically significant differences inthe presence of HER-2(+)69.6%and HER-2(-)33.3%were noted in the expression rate ofTopoIIα in infiltrating ductal breast carcinoma tissues (P<0.01). In the group of Ki-67(+),the positive expression rate of TopoIIα was68.3%, while it was27.3%in the Ki-67(-)group, which has statistical significant differences. No statistically significant differenceswere found in infiltrating ductal breast carcinoma tissues between Topo Ⅱα and age, tumorsize, histological class, clinical stage, ER or PR (P>0.05). Conclusions: The higher expression rate of TopoIIα in breast carcinoma was associatedwith lymph node metastases, positive expression of HER-2and Ki-67, while there was nodirect relationship with age, tumor size, histological class, clinical stage, or expression ofER and PR. These results suggest that the increased expression of TopoIIα may play animportant role in the development and progression of breast carcinoma, which can be usedas indicators of clinical treatment and prognosis.
