ObjectiveTo study whether stabilizing microtubules could prevents myocardial mitochondrial ischemia-reperfusion injury.Methods1. The ischemic ventricular arrhythmia model was established by using isolated rat heart langendorff-perfused model. They were divided into five groups(each group=15), control group, ischemic group, ischemic+taxol (0.1μM) group, ischemic+taxol(0.3μM) group and ischemic+taxol(1μM) group. After a20-min equilibration period as follows:control group,150min normoxic superfusion; ischemia group,30min ischemia,120min normoxic superfusion; Taxol group,30min ischemia,120min normoxic superfusion, with Taxol at a concentration of0.1(0.3and1μM) throughout the entire150-min superfusion period. And arrthymias scores were assessed.After perfusion, left ventricular muscle were taken to make frozen sections. Microtubules were observed by immunohistochemical methods; Kits were used for detecting the quantity of reactive oxygen species, as well as oxidase activity detection by fluorescence photometer and spectrophotometer to detect. Results1.Taxol (1μM) signifycantly reduced the infarct size as compared with control (P<0.05).2. Further study revealed that0.1,0.3and1μM Taxol reduced the level of ROS by33%,46%and51%, respectively (P<0.05).3. 0.3and1μM Taxol increased the activity of mitochondrial electron transport chain complex I, whereas0.1,0.3and1μM Taxol increased that of mitochondrial electron transport chain complex Ⅲ.conclusionStabilizing microtubules could prevents myocardial mitochondrial ischemia-reperfusion injury.
