BackgroundIt is presume that the cause of recurrent hydatidiform mole is NLRP7gene by studying familial hydatidiform mole. Its possible mechanism is thought as inflammation pathway. hydatidiform mole is benign gestational trophoblastic tumor(GTT),but is the GTT which from hydatidiform mole associated with the NLRP7gene is uncertain.ObjectiveThis research was detecting NLRP7protein, caspase-1protein,pro-IL-1β protein, and expression of cytokines IL-1β by peripheral blood lymphocytes culture of the GTT women and normal control of women.Materials and methodsSelecet10patients and10controls. The patients including10GTT patients.as well as choice10cases of women as a control whose growth history is normal with no adverse reproductive outcomes. The major process is taking their peripheral blood15-20ml, lymphocytes culturing respectively by LPS stimulation. collecting culture fluid for the determination of IL-1beta, collecting cells to Western Blotting for determination of NLRP7Caspase-1and pro-IL-1β expression and comparing the2groups.Results(1) IL-1β secreted cytokinesIt can release the data analysis of IL-1β secreted cytokines:after LPS stimulation, secreted group IL-1beta cells were slightly lower than that of the control group.2cases had obvious difference (Grade11,,55)(2) the expression of NLRP7protein and caspase-1protein:By Western blotting in6pairs of NLRP7and caspase-1protein results obtained: NLRP7expression had no difference, but2cases of48kd casepase-1protein have no expression in case group (Grade11,55).(3) the expression of pro-IL-1p proteinby Western blotting2pairs of pro-IL-1beta protein expression Results obtained: the expression of pro-IL-1β protein in case group was higher than that in control groupConclusionsNo48kd casepase-1expression in gestational trophoblastic tumor which from hydatidiform mole, may be one of the possible causes of casepase-1dependent low IL-1β secretory...
