Objective:To investigate the effects of FuFang Danshen Injection on he expression ofNO、bcl-2and S100B which were from septic shock rats with brain injury.Methods:Forty2-week-old healthy Wistar rats were randomly divided into four groups.normal group(n=10):This group was detected blood pressure by ECG and was giventhe same amount of normal saline;septic shock group1(6h)and septic shock group2(12h):The two groups were injected lipopoly saccharide (5mg.kg-1) through thetail vein to make the septic shock model and the phenomenon that blood pressure dr-opped to the original2/3, with restlessness and skin cyanosis indicatesd successfulmodeling;Fufang Danshen Injection treatment group:This group was intraperito-neally injected Danshen injection (5ml.kg-1) immediately when shock signsappe-ared.Except septic shock group2which was drawn12h after the successful modeli-n g,the remaining three groups were harvested6h after the success of the model.TheNO content in hippocampal organization was detected withith nitrate reductase way,the excpresion of Bcl-2in hippocampal tiusse were investiged with immunohistoch-emistry technique,meanwhile the serum concentr-ations of S100B were measured byenzyme--linked immunosorb-entassay (ELISA).Results:1.In brain tissue NO content determination:the content of NO from Septic shockgroup1(6h) was higher than that from the normal group (P <0.05);compared with theseptic shock group1,the content of NO from the treatment group was significantlylower (P <0.05), but the septic shock group2NO content was higher than septic shockgroup1(P <0.05).2.Hippocampal tissue expression of bcl-2:Normal group, septic shock group1(6h)and treatment group were significantly different between groups(P <0.05). Compared with normal group, the expression of bcl-2in septic shock group1and treatmentgroup was significantly increased(P <0.05),meanwhile the treatment group bcl-2expression increased than that in the septic shock group expression(P <0.05);but theseptic shock group2bcl-2expression was lower that in septic shock group1(6h)(P<0.05).3.The content of serum S100B:Septic shock group1S100B was higher than thatin the normal group (P<0.05),and compared with the septic shock group,the contentof S100B in the reatment group decreased significantly (P<0.05),but the septic shockgroup2(12h) of S100B was higher than that from septic shock group1(P <0.05).4.Histopathological changes in the brain:Normal control group: Level section ofbrain cells was clear and the structure was complete,nerve cells with abundantcytoplasm and nucleolus were observed;Septic shock group1and Septic shock group-2: The normal structure disappeared or was less clear, nerve cells disorganized andconfused, and some cell necrosis; Treatm ent group:normal tissue structures weresurviving in and interstitial edema was mild,nerve cells arranged regularly, karyopyknosis was less and the structure was more complete.5.NO content in brain tissue and bcl-2in hippocampus relevant analysis:there wasa negative correlation between the NO content in brain tissue and bcl-2in hippocam-pus (r=-0.928, P<0.05).6.NO content in brain tissue and serum S100B levels correlation analysis:therewas a positive correlation between NO content in brain tissue and serum S100Blevels(r=0.877, P <0.05).Conclusion:1.Excessive NO can cause apoptosis of brain cells during septic shock and themechanism may be related to inhibition of bcl-2expression.2.NO and S100B are involved in the process of brain injury during septic shock.3.The expression of S100B can reflect the extent of brain cell damage duringseptic shock.4.FuFang Danshen Injection has a protective effect on rats brain tissue during septic shock, and its mechanism maybe include reducing the content of NO andelevating the expressions of bcl-2.
