Background and objective: So far,the pathogenesis of hepatic encephalop-athyis not clear and have not clear treatment.that It has been the object of theclinical medicine integrated with Chinese traditional medicine and Westernmedicine.the mortality of Hepatic Encephalopathy of type A (AHE) is up to80%which is common in critically ill disease.Xingnaojing injection can shortenthe time AHE coma,promote sober,and have clear clinical efficacy.However, thedata of clinical efficacy and clinical case are very limited,which having noevidence to support the treatment of xingnaojing for AHE.In this study,we investigate the optimal induction time of Thioacetamide (TAA) for AHE model in rat.Study the effect of Xingnaojing on hepatic functionendotoxin,the expression of Glutamine Synthetase (GS) and Nitric OxideSynthase(NOS).The study provides a scientific evidence for the AHEtreatment of Xingnaojing.Method:1Studying the optimal induction time of Thioacetamide (TAA) for AHEmodel in rat:(1) The60rats were divided into four groups which Include B, C, D.The groups of A is Normal control group with gavaged by equal amount ofphysiological saline for4days.The group of B, C, D is the experimental group respectively with TAA (300mg·kg-1·d-1)2days,3days and4daysthrough intragastric administration.(2) Comparing the behavioral changes of rat,the induction rate and Mortal-ity of rat with AHE;Analyzing the difference of blood ammonia Glutamic Ox-alacetic Transaminase(AST), Glutamic-Pyruvic Transaminase(ALT) and Total Bilirubin(TBil) after24hour;Comparing the morphological each group;Comparing the morphological changes of liver tissueineach group;Analyzing the content of endotoxin,INF-α,and The expression ofGS and NOS.Results:1The optimal induction time of Thioacetamide (TAA) for AHE model inrat:(1) The brain function classification of the group of B,C,D is more thanthe group of A(P<0.0083).The brain function classification of the group of C,Dis more than the group of B(P<0.0083).The difference have statisticallysignificant.(2) A group without rats induced by AHE and death;The induction rateof the group of C,D is more than the group of B (P<0.0083).The differencehave statistically significant.The fatality rate of the group of D is more thanthe group of B,C (P<0.0083).he difference have statistically significant.(3) The levels of ammonia,AST,ALT,TBIL of the group of B,C,D is more than the group of A(P<0.05).he difference have statistically significant.The levels of ammonia,AST,ALT,TBil of the group of C,D is more than thegroup of B(P<0.05).The difference have statistically significant.(4) The histomorphology of liver of A group is normal.Inflammatory Infilt-ration,necrosis,and fibrosis in of the group of C,D in live tissue damagedobviously.2Study on the effect of Xingnaojing on hepatic function, endotoxin,theexpression of GS and NOS:(1) The histomorphology of liver in A group is normal;The brain functionclassification of the group of C,D,E,F is more than the group of B(P<0.0033).The difference have statistically significant.(2) The levels of ammonia,AST,ALT,TBIL of the group of B,C,D,E,F ismore than the group of A(P<0.05).The difference have statistically significant;the levels of ammonia,AST,ALT,TBil of the group of C,D,E,F is more thanthe group of B(P<0.05).The difference have statistically significant;The levelsof ammonia of the E group is lower than C group(P<0.05).The differencehave statistically significant;(3) The histomorphology of liver of A group is normal;the morphologicalchanges of liver tissue in E group is lighter than the group of C,D,F.The Egroup showed a small amount of inflammatory infiltration, spotty necrosis, andpartial liver cell swelling;(4) The levels of endotoxin ane INF-α of the group of B,C,D,E,F is morethan the group of A(P<0.05).The difference have statistically significant;Thelevels of endotoxin of E group is more than the group of C,D(P<0.05).Thedifference have statistically significant;(5) the expression of GS and NOS of the group of B,C,D,E,F is morethan the group of A(P<0.05).The difference have statistically significant;theexpression of GS of the group of C,D is more than the group of E(P<0.05).The difference have statistically significant;the expression of GS and NOS ofthe group of B,C,D,E,F is more than the group of B(P<0.05).The difference have statistically significant;the expression of NOS of the group of D,E,F ismore than the group of C(P<0.05).The difference have statistically significant.Conclusion:1The appropriate time induced AHE in rats is TAA(300mg·kg-1·d-1) forfour days by gavage.This method has significantly behavioral changes in rats,lower death rate, high blood ammonia, and Significant liver damage.2Xingnaojing can improve the behavioral changes, inflammatoryinfiltration,necrosis, reduce the level of iendotoxin and INF-α,and lower theexpression of GS and NOS.In the treatment of different doses of Xingnaojingand L-Ornithine L-aspartate salt,xingnaojing(10ml·kg-1·d-1) has better comprehensive effect inthe treatment of AHE.
