Purpose:To observe Quinoline derivatives act on PC3prostate cancer cells of intercellular gapjunctions, from the level of cell communication further explore intercellular gap junction effectby cisplatin drug. To study the effect of the combined treatment of prostate cancer cells andthe possible mechanism.Materials and methods:Use prostate cancer cell line PC-3as the research object, add differentconcentration of Quinoline and cisplatin into postate cancer cell line PC-3,and divided intogroups,24hours and48hours two time points. CCK8method was applied to detect cell’svitality, real-time PCR method to detect the expression of cell gap junction associated proteinCx43mRNA and Western Blot to detect the expression of Cx43protein. Application of laserconfocal detection cell GJIC function changes.Results: CCK8method detection results showed that in different concentration of quinolinederivatives in combination with cisplatin affects PC3cells after24,48h, PC3prostate cancercells more active in cisplatin group is obviously different. Real-time PCR and Western Blot testresults showed that different concentrations of quinoline in combination with cisplatin acts onPC3cells after24h, Cx43mRNA and protein have varying degrees of increase. Compare the useof cisplatin alone Cx43mRNA and protein expression level difference has significant statisticalsignificance (P <0.05). Confocal laser experiment results show that the PC3prostate cancercell GJIC function with varying degrees of enhancement, with drug concentration increased,the number of fluorescent dyes in the expression of cell gap junction, fluorescence intensityalso increase, and the dose effect dependence and time dependence effect of dependencies.Conclusion: Quinoline can increase prostate cancer PC3cells intercellular gap junction Cx43gene mRNA and protein expression level and promote the PC3prostate cancer cell GJICfunction.When combine with cisplatin, it also can enhance the effect of cisplatin on PC3prostate cells.It is possible to provide a new method of combined treatment of advancedprostate cancer.
