Purpose:used prostate cancer cell line PC-3as the research object,to discuss the influence of resveratrol and cisplatin combined actionon proliferation of hormone independent prostate carcinoma cells andthe relative possible mechanism.Methods:add different concentration of resveratrol and platinumin prostate cancer cell line PC-3,and divided into5groups asfollow(group(1):used different concentration of resveratrol separately effect for24h;(2)used different concentrationsof cisplatin separately effect for24h;(3)the different concentration of cisplatin and5mmol/L of resveratrol effect atthe same time for24h;(4)1mmol/L,5mmol/L resveratrol effectfor24h,added different concentrations of cisplatin,and continue cultured for24h;(5)24h after the different concentration of cisplatineffect,add1mmol/L,5mmol/L resveratrol,continuecultured for24h;)used Flow cytometry to detect cell cyclephase,and tested the gene expression and protein expression ofLC3,P glycoprotein and multidrug resistance related proteins and mTOR,comparedthe influence of each group on PC-3cell proliferation.Results:1. Resveratrol and cisplatin alone,inhibits prostate cancer cellline PC-3cell proliferation,in the certain concentration range was doseassociated;2.the joint action of resveratrol and cisplatin can inhibit the growthof prostate cancer cell line PC-3,the effect is more better than themedicine group,and the cell block in G2,inducing cell apoptosis.Cisplatin and the joint action of resveratrol effect on cell proliferation,relatedto the medication order.3. RT-PCR and Western blot results show that under the action ofresveratrol effect alone or in combination with cisplatin plus,multi-drugresistance protein MRP,protein P glycoprotein,amount of gene and proteinexpression of mTOR inhibitors are reduced.The increased amount of geneand protein expression of LC3.Resveratrol and cisplatin combination groupof LC3,mTOR inhibitors,multi-drug resistance related proteins,protein Pglycoprotein gene and protein expression were higher than the medicinegroup,the differences between the groups was statistically significant(P <0.05).Conclusion: Resveratrol and cisplatin combination can be enhanced forprostate cancer cell line PC-3proliferation inhibition, and isassociated with both way of medication, the related mechanism of actionand affect the cell cycle distribution and apoptosis induced and regulatedrug resistance related proteins.
