Objective:Recent studies show that human male fertilities have been increasinglyimpaired due to a variety of factors, such as genetic and environmental factor. Theincidence of infertility and reproductive system abnormality was significantlyincreased. Cryptorchidism is one of the most common diseases in human malereproductive system congenital malformation. Cryptorchidism has high incidence, andits long-term major complications include: adult infertility and an increased incidenceof testicular cancer. The studies on mice worldwide prove that INSL3and LGR8arethe main regulating factors for testicular growth and development and play a decisiverole in the process of testicular descent. The thesis studies the mRNA expression ofNSL3and LGR8in hydrocele and testicular gubernaculums of a child with unilateralcryptorchidism and testicular hydrocele, and further revealed the relevance betweenNSL3and LGR8and the incidence of cryptorchidism.Methods: By RT-PCR, compare the mRNA transcriptions of INSL3and LGR8in thetunica vaginalis testis and gubernaculums testis in42children patients with unilateralcryptorchidismand40non-cryptorchid children in a similar age。Results:①The mRNA expression level of INSL3and LGR8in the cryptorchidismhydrocele of a child with cryptorchidism is lower than in a normal children of asimilar age (P<0.05). This suggests that the INSL3and LGR8gene expression levelin children with cryptorchidism may be generally low. In the present study, the lowexpression of INSL3and LGR8in hydrocele and testicular gubernaculums suggestsincomplete male differentiation.②The expression level of mRNA of INSL3and LGR8in cryptorchid gubernaculumsof a child with cryptorchidism significantly lower than their expression incryptorchidism sheath, and even a single sample nealy has no expression. It suggeststhat INSL3and LGR8may regulate the growth and development of human testiculargubernaculums, and play a decisive role in the process of testicular descent. Theabnormal low expression of INSL3and LGR8might be one of the causes of humancryptorchidism.Conclusions:The mRNA expression of INSL3and LGR8in the hydrocele andgubernaculums testis of the cryptorchidism testes in a child with unilateralcryptorchidism decreases to a lower level, especially to a significantly low level in thegubernaculums testis. The INSL3and LGR8gene expression level in cryptorchidismchildren’s reproductive system may be generally low, and cryptorchidism is an incomplete differentiation of maple differentiation INSL3and LGR8may regulate thegrowth and development of human testicular gubernaculums, and play a decisive rolein the process of testicular descent. The abnormal low expression of INSL3and LGR8might be one of the causes of human cryptorchidism.
