Objective:To assess the effect of hypoxia on apoptosis of lung tissue by observingemphysema rat lung tissue Caspase-3expression level at different altitudes.Methods:40wistar rats were randomly divided into Comparison Group, MediumAltitude Saline Group, Medium Altitude Emphysema Group, High Altitude SalineGroup, High Altitude Pulmonary emphysema group, n﹦8. Rats of Comparison Group,Medium Altitude Saline Group and Medium Altitude Emphysema Group were fed inQinghai University Medical School animal room at2260m altitude, by day and night,light and airy, the room temperature is about25℃, regularly disinfected, the ratswere free feeding and drinking water. High Altitude Saline Group and High AltitudePulmonary emphysema group were raised in low pressure oxygen tank that at highaltitude Qinghai University Medical Center School of Medicine, the low pressureoxygen tank condition is: simulated5000m altitude, pressure:397.5mmHg, oxygenpartial pressure:52889Pa~53001Pa, temperature:16.9℃-24℃, humidity:37.3%-51%. Animal’s feeding, drinking, drug injection, cleaning and disinfectionwere carried out in the low pressure oxygen tank. The rats of Comparison Group didnot receive any treatment, Rats of Medium Altitude Saline Group and High AltitudeSaline Group were intraperitoneal injected with saline0.8ml on day1,8days,15days,22days, Medium Altitude Emphysema Group and High Altitude Pulmonaryemphysema group were injected at the same time with cigarette smoke extract(Cigarette Smoke Extract, CSE) solution0.8ml in their abdomens. After4weeks’feeding, immunity group collected the right lungs of the rats and did chemical test tocheck the expression of Caspase-3of the lung issue.Results:Compared with Comparison Group and group Medium Altitude SalineGroup、High Altitude Saline Group, Medium Altitude Emphysema Group and HighAltitude Pulmonary emphysema group parenchymal damage, some alveolar ruptureand alveolar expansion in rats by intraperitoneal injection of CSE successfully established emphysema model.Compared with Comparison Group、Medium Altitude Saline Group andHigh Altitude Saline Group, lungs of rats of Medium Altitude Emphysema Groupand High Altitude Pulmonary emphysema group were almost damaged and part of thealveoli broke and alveolar space expanded. The results showed Caspase-3cellpositive rate varies significantly among the5groups (P <0.001). Cell positive rate ofHigh Altitude Saline Group (11.63%±0.81%)was higher than Comparison Group(3.67%±0.49%)and Medium Altitude Saline Group(3.97%±0.94%)(P <0.001), High Altitude Pulmonary emphysema group (19.33%±0.92%)was higherthan Medium Altitude Emphysema Group(10.71%±1.07%)(P <0.001), MediumAltitude Emphysema Group was higher than that of Medium Altitude Saline Group (P<0.001), High Altitude Pulmonary emphysema group was higher than that of HighAltitude Saline Group (P <0.001).Immunohistochemical results observed under40×10magnificationmicroscope tips, Caspase-3positive in lung tissue was significantly increased in HighAltitude Pulmonary emphysema group, Medium Altitude Emphysema Group andHigh Altitude Saline Group reduced the number of Caspase-3-positive groupcompared with High Altitude Pulmonary emphysema group, Comparison Group andMedium Altitude Saline Group expression is minimal.Conclusion:Caspase-3cell positive rate of high-altitude pulmonary emphysema inrats increased, The plateau low oxygen accelerated apoptosis expression of rats withemphysema.
