Objective To study the influence of hypobaric hypoxia on the lung architecture in Ratswith emphysema, we measured mean linear intercept(MLI), destructive index(DI),area of pulmonary alveoli (APA) and Density of pulmonary alveoli(DPA).Methods Wistar(n=40)rats were randomly divided into five groups:(1)blankcontrol group in moderate altitude,(2)saline group in intermediatealtitude,(3)emphysema group in intermediate altitude,(4)saline group and (5)emphysema group in high altitude. The low pressure oxygen tank condition is:simulated5000m altitude, pressure:397.5mmHg, oxygen partial pressure:52889Pa~53001Pa, temperature:16.9℃-24℃, humidity:37.3%-51%. CSE and0.8ml of salinewere injected in emphysema and saline groups intraperitoneally once a week for fourweeks. After HE staining,we measured MLI, DI, APA and DPA.Results The MLI (189.22±27.96umvs.158.30±28.37um), DI (53.34±6.54%vs.20.90±3.57%) and APA (5740.12±749.25um2vs.3101.25±763.43um2) weresignificantly increased in the emphysema group compared with saline group inintermediate altitude group (p<0.001, respectively). However, the DAP(184.53±30.18mm2vs.334.02±50.02mm2, p<0.001) was significantly decreased inthe emphysema group rats. The MLI (226.61±27.51um vs.193.13±24.66um), DI(73.98±6.97%vs.57.64±6.72%) and APA (10606.83±923.057um2vs.5399.42±1058.47um2) in emphysema group were higher than in saline group rats inhigh altitude (p<0.001, respectively), DAP (88.84±5.67mm2) was lower than the latter(182.11±39.79mm2, p<0.001). The saline group of the value of MLI, DI and APA inhigh altitude were higher than in intermediate altitude (p<0.001, respectively), DPAwas lower than the latter (p<0.001). The MLI, DI and APA in emphysema group weresignificantly increased in high altitude compared with in intermediate altitude group(p<0.001, respectively). The DAP was significantly decreased in high altitudeemphysema group rats.Conclusion The indices of MLI, DI and APA with emphysema groups in highaltitude were higher than intermediate altitude, DAP significantly reduced, that thehypobaric hypoxia of altitude environment aggravates the damage of the Lungarchitecture in rats with emphysema.
