Objective:Objective:To investigate the expression of miR-127、miR-155and miR-200a in sporadic Burkitt’s lymphomas (BL) in children.with positive andnegative Epstein-Barr virus(EBV) infection.Methods:1、40case of BL and15case of tonsil tissue samples were collectedfrom hospitalized patients of Department in Children Hospital of Jiangxi Proviencefrom2005to2012.To analyze the clinical characterics and prognosis of BL, byusing tissue chip and ISH to test the expression of EBV in BL. According to EBVgenetic test results, BL was divided into into EBV-negative group, EBV-positivegroup and normal control group.2、qRT-PCR was used to detect the expression of miR-127、miR-155andmiR-200a in these case,including40case of BL and15case of tonsil tissue.3、qRT-PCR was used to detect the expression of miR-127、miR-155andmiR-200a in Raji and Ramos.4、FISH was used to detect the expression of c-myc in these case,including40case of BL and two BL cells line.5、SPSS13.0software was used for statistical analysis of the experimentaldata.Measurement data were in the form of “mean±standard deviation”, Countdata using t test. P <0.05was considered statistically significantResults:1、16cases of BL EBV-positive (40%), in these cases2patients(12.5%) located in the mesenteric lymph nodes,4cases (25%) located in theileocecal intestinal tissue,10cases (62.5%) located in the mesenteric+ileocecal;24cases of BL EBV-negative (60%), in these cases,2cases (8.33%) is located in themesenteric lymph nodes,6cases (25%) located in the ileocecal intestinal tissue,16cases (66.67%) is located in the mesenteric+ileocecal.15cases of tonsil tissueEBV-negative (100%)2、c-myc gene translocation was38cases(95%), of which EBV-negative was23cases (60.53%) and EBV-positive was15cases (39.47%);2cases of BL havenormal c-myc gene (5%), of which EBV-positive was1case and EBV-negative was 1case (50%each). Raji c-myc gene translocation positive, Ramos have normalc-myc gene.3、Compared EBV-positive BL with EBV-negative BL cases and tonsil tissuemiR-127expression was down-regulated(P﹤0.05). Compared Raji with Ramos,miR-127expression was down-regulated(P﹤0.05).The expression of miR-155inEBV-positive and EBV-negative BL had no significant differences(P>0.05),.Theexpression of miR-155in BL compared with tonsil tissue was overexpressed(P﹤0.05). Compared Raji with Ramos, miR-155expression was down-regulated.Compared EBV-positive BL with EBV-negative BL cases and tonsil tissuemiR-200a expression was up-regulated(P﹤0.05). Compared Raji with Ramos,miR-200a expression was up-regulated(P﹤0.05).The expression of miR-127、miR-155and miR-200a had no significant correlation with c-myc gene(P>0.05).Conclusion:1、 The expression of miR-127、 miR-155and miR-200a was different inEBV-positive and EBV-negative in sporadic Burkitt’s lymphomas (BL) in children.2、Expression of miR-127、miR-155and miR-200a is closely related to thepathogenesis of EBV-associated BL.
