Taking the edible fungi Taishan Boletus edulis as material, this research established the methods for the quantitative determination of Boletus edulis Polysaccharides (BPS), furthermore investigated the optimum technological parameters of extracting conditions for BPS, then indicated the separation and purification technology of BPS crude samples, and stuided Taishan Boletus edulis crude polysaccharide antioxidant function and anti-aging function finally. The conclusions of this research were as follows:1 The methods for the quantitative determination of active components polysaccharide in Boletus edulis were established. The content of total sugar was phenol- oil of vitriol mensuration, while the content of reductive sugar was 3, 5-Dinitrosalicylic Acid mensuration, however the content of Boletus edulis Polysaccharides depended on their concentration difference.2 The optimum technological parameters of three extracting conditions for BPS were compared. At first, traditional hot water extraction for BPS were studied, which the optimum technological conditions were disintegrating the dry powder by 80 granularity sieve, the ratio of water to solid was 40, extracting at 100℃for 3 hours twice respectively, vacuum concentrated to proper volume and then precipitated with 3 times volume of 95% ethanol ,and the average final content of BPS was 4.29%. Microwave extraction for BPS were studied, which the optimum technological conditions were reaction time 55 seconds, ratio of material to water 1:40,microwave power 640W, the average final content of BPS was 5.69%. Ultrasonic extraction for BPS were studied, which the optimum technological conditions were reaction temperature is 50℃, reaction time is 40 minutes, ratio of material to water 1:40, the average final content of BPS was 4.83%.3 The separation and purification technology of BPS crude samples. The extracting solution of BPS concentrated by vacuum rotary evaporator, then eliminated protein using sevag reagent and dialysed by distilled water. When ethanol of certain concentration added, many brown deposit were formed. Then the deposit dried by vacuum freeze drier. Above all, purification was achieved by DEAE-52 Ion Exchange Chromatography. Furthermore the grads elution diagrams of the BPS samples on Sephadex G-200 Gel Chromatography were also received. In the end, the separated compositions physicochemical characters and structural Properties were analysed.4 The antioxidant function and anti-aging function of BPS crude samples. Mice were infused with different concentrations BPS Solutions, Infusing time continued for a month. Mice weight and serum SOD, MDA and GSH-PX were indicated. The experimental results showed that: Taishan Boletus crude polysaccharide could strengthen the effect of antioxidant function. Glucose, BUN, and LDH were indicated. The experimental results showed that: Taishan Boletus crude polysaccharide could strengthen the effect of anti-aging function.
