Floccularia luteovirens is famous as a medicinal fungi.Not only it has the efficacy ofanti-tuumor, anti-aging, prolong life, such as skin care, beauty effect, at the same time,becauseof the high conent of iron, make it the ideal blood recuperate healh food and energy metabolism,enhance skin resisance, prevent the functional food. For the further discovery the value ofFloccularia luteovirens, the experiment was observed the effects of FLP, alcohol extracts andthe polar parts of Floccularia luteovirens on human breast MCF-7and ovarian SKOV3.Andtested on normal mice and mice treated with H22separately. Using the traditional method ofwater extract-alcohol precipitation, Floccularia luteovirens polysaccharide(FLP) was extracted;Selecting FLP as experimental material, detected the regulation of FLP to the four kinds ofcytokines: CRP, INF-γ, TNF-α and IL-6in serum. Determined the effects of FLP on theexpression of the gene of the intestinal samples and the immune function in normal mice byReal-time quantitative polymerase chain reaction(RT-qPCR). Detected the regulation of FLP tothe four kinds of cytokines: CRP, INF-γ, TNF-α and IL-6in serum in mice treated with H22.The results were as follows:1、By using high-performance liquid chromatography(HPLC), measured FLP sampleβ-glucan and mannan oligosaccharide content of20.1%and5.7%. Further experiments exploredthe anti-tomor effect and immunomodulatory effect of Floccularia Luteovirens.2、For more comprehensive and further evaluating and developing Floccularia luteovirens,on the basis of previous experiments what showed that the active components in petroleum etherand ethyl acetate layer of it has a high cytotoxic effect on human ovarian cancer HO-8910.Accordingly, this paper filtered the effective component of which had the effect of antitumor ofFloccularia luteovirens fruilting body on human breast MCF-7and ovarian SKOV3. Detectedthe promoting apoptosis effect of95%ethanol extract,75%ethanol extract, macroporous resinaqueous extract(water level), petroleum ether, ethyl acetate and normal butanol layer extractsand FLP on human breast MCF-7and ovarian SKOV3. Experiment results showed that, thepromoting apotosis effect of each experimental group on human breast MCF-7in descendingorder is: ethyl acetate layer extracts (34.54%)> petroleum ether layer extracts (32.21%)>75%ethanol extract (27.06%)> FLP extracts (26.03%)>95%ethanol extract (23.71%)> normalbutanol layer extracts (20.10%); Correspondingly, the optimal concentrations were ethyl acetatelayer extracts (100μg/mL), petroleum ether layer extracts (100μg/mL),75%ethanol extract(100μg/mL), FLP extracts (200μg/mL),95%ethanol extract(50μg/mL), normal butanollayer extracts(50μg/mL); The promoting apotosis effect of each experimental group on humanovarian SKOV3in descending order is: ethyl acetate layer extracts (60.80%)> petroleum etherlayer extracts (51.64%)>75%ethanol extract(42.96%)>95%ethanol extract(40.85%)> FLPextracts (35.45%)> normal butanol layer extracts(30.52%); Correspondingly, the optimalconcentrations were ethyl acetate layer extracts (400μg/mL), petroleum ether layer extracts(400μg/mL),75%ethanol extract(100μg/mL),95%ethanol extract(200μg/mL), FLP extracts(100μg/mL), normal butanol layer extracts (200μg/mL).3、Studied on the antitumor effect of FLP on mice treated with H22, detected theregulation to the four kinds of cytokines: CRP, INF-γ, TNF-α and IL-6in serum. Analysed theimmune organs index, the regulation of FLP to the four kinds of cytokines: CRP, INF-γ, TNF-αand IL-6in serum and discussed the mechanism to antitumor effect. Established the model ofmice treated with H22, calculated the weight of tumor, thymus index and spleen index,measured the levels of CRP, INF-γ, TNF-α and IL-6. Experiment results showed that, the tumorinhibition rate of each experimental group in descending order is: the CTX group> FLP-Mgroup> FLP-H group> FLP-L group> the control group; All the dose group showed theantitumor effect varying degrees of FLP, improved the immune organs index and increased theconcentration of CRP, INF-γ, TNF-α and IL-6in serum. According the results,the antitumoreffect of FLP was likely to reach by humoral regulation.4、Determined the immune function of FLP on normal mice.In this experiment, fed miceon FLP by health care products, texted the immune organs index, the regulation of FLP to thefour kinds of cytokines: CRP, INF-γ, TNF-α and IL-6in serum, the expression of the gene of theintestinal samples and the immune function in normal mice. Discussed the mechanism toimprove the immune function. Experiment results showed that, compared with the control group,FLP can significantly improve the immune organs index, decrease the level of CRP and increasethe concentration of INF-γ, TNF-α, IL-6in serum. The expression of the gene of the intesinalsamples increased in varying degrees, showed significant difference to what compared with thecontrol group. According the results, FLP promote humor regulation by collaboration the genesregulating the body’s immune function, in order to realize the immunomodulatory effects.
