Bulgaria inquinans (Pers.) Fr. is a special fungus with photosensitive activity, and it’spotential medicinal value has been taked as the target of fungi researchers. Bulgaria inquinans isthe edible fungi. People in the Changbai Mountain area always simply process the fruitingbodies of collected Bulgaria inquinans, in order to avoid the occurrence of light allergicdermatitis reactions.The chemical composition of Bulgaria inquinans is rich, which contain a lot of ergosterol,quinones, and phenylpropanoid compounds etc. Some characteristical active substances areincluded, for example, benzo[j]fluoranthene class, azaphilone class, perylenequinonoid andothers. The components of azaphilone class have excellent antibacterial activities, and theperylenequinones also have multiple pharmacological activities. These secondary metabolitesenrich the components contained in Bulgaria inquinans, while make it possible to be as amedicinal mushroom with more multi-development direction.Based on previous study on Bulgaria inquinans, the separation and identify of chemicalcomposition from the fruiting bodies were further researched in this paper.4,9-dihydroxy-1,2,11,12-tetrahydroperylene-3,10-quinone was obtained from petroleum etherextract, the work emphasis on the colorful component portion separated of dichloromethaneextracts was extracted, to give3β,5α-dihydroxy-(22E,24R)-ergosta-7,22-dien-6-one,bulgareone A,1,3,6,8-tetrahydroxyanthraquinone,1,3,8-trihydroxy-6-methylanthracene-9,10-dione, ergosterol, mannitol.In photosensitive activity of Bulgaria inquinans fruiting bodies, the photosensitive activitycomponent was tracked through the active tracking method. The portion containedphotosensitive active ingredient is identified in acetone extract. And after according to polaritygradient extraction, the photosensitive active ingredient having a concentrated in methylenechloride extract, while petroleum ether extract and ethyl acetate extract also photosensitiveactivity, but the experiment testified that the content of activity components is extremely lowthrough coarse points cut section processing. A series of methods were taked, which includedextraction and separation to the mixtures of methylene chloride extract, petroleum ether extractand ethyl acetate extract, mouse tracking the active experiments, taking ear and head swellingafter administration as standard, silica gel column chromatography, LH-20gel columnchromatography, high performance liquid chromatography (HPLC), recrystallization, etc. In the end, the non-photosensitive active impurities were excluded and the photosensitive activefraction was containied, but photosensitive active substance structure is very unstable, theconventional method is difficult to obtain the enough pure compound which could be identified,a new means of separation or other means are expected to obtain the ingredients.The photosensitive active component with certain purity, obtained in the above experiment,was feeding in the gavage mice experiment. Result was found that after light clearlyphotosensitive reaction was on the skin, head and fewer fur part limbs of mice. Ear swellingphenomenon is the most obvious, it is certain that the active ingredient mainly gathered inphotosensitive reaction areas such as the mice ears. Therefore, in order to further determine thephotosensitive active composition, put to death in mice after appearring photosensitive response,cut off the ear, take the blood, liver, kidney and spleen, extract with organic solvent, theextraction was analyzed by GC-MS, preliminary findings is that medicine group of mice earhave significant difference with the blank group.The photosensitive active ingredient was isolated and purified through preparative HPLCfrom methylene chloride extract. Although there was a single absorption peak at440nm, no purephotosensitive active substance was finally obtained. Failure to obtain the photosensitive activemonomer may be due to the physicochemical instability of the photosensitive active ingredient.One main ion peak of453M were obtained by HPLC tandem mass analysis. We can speculatethat the molecular weight of the photosensitive active ingredient is454M.It showed differences of ears, liver, kidney through comparison between the administeredmice with the control group by HPLC analysis. Separate the components, two components wereobtained from the extract of administered mice ears and two components from liver and kidney.The four samples were analyzed by MS, a component of molecular weight close to get positivespectrum of560M,514M negative spectrum of ingredients, was contained in the extracts of themice administered ears, liver, kidney. So combined with the obtained results of gaschromatography-tandem mass, it suggest that compound may be sensitive active ingredients orthe metabolism conversion product in the body.
