Background and Objectives:The relationship between p53gene codon72polymorphism and risk of cancer had been investigated in many typesof tumor, however the results remain controversy. The aim of presentstudy is to investigate the association of the single nucleotidepolymorphism (SNP) of codon72of p53gene (Arg72Pro) and risk ofcolorectal cancer in QingHai area of China.Materials and Methods:147cases of colorectal cancer patients and146healthy controls were collected. Genotyping was performed withreal-time polymerase chain-restriction fragment length polymorphism(PCR-RFLP). Genotype frequencies were compared between patients andcontrols, and the relationship between genotypes andclinicopathologicaldata was analyzed.Results: The frequencies of Arg/Arg, Arg/Pro, and Pro/Pro genotypes ofthe p53codon72polymorphism were33%,46%and21%in thecolorectal cancer patients; and44%,46%and10%in the health controls,the difference between two groups were significant (P=0.024).Stratification analysis indicated that pathologic differentiation, but notage, gender, nation and TNM stage, was associated withgenotypes,Arg/Arggenotype patients with more well-differentiated adenocarcinoma and Pro/Progenotype patients with morepoor-differentiated adenocarcinoma.Conclusions:p53Prohomozygosity isassociated with the increased riskof colorectal cancer in QingHai area of China, and associated withpoor-differentiatedadenocarcinoma as well.
