Objective:To detect the the expression and significance of chemokine receptor5inperipheral blood CD4+CD25+CD127-regulatory T rells in patients with gastric cancerand healthy controls. To explore its relationship with the occurrence, invasion andmetastasis of gastric cancer.Methods:Using flow cytometry technique(FCM) to detect the expression level ofCCR5in CD4+CD25+CD127-regulatory T rells in49gastric cancer patients and20healthy controls peripheral blood and to study the relationship of the expressionlevel with clinical pathological parameters. SPSS19.0was used for stastistcal analysi.T test and One-way ANOVA were used for measurement data. Linear Regressssionand Spearman test was used for association between group.Results:(1)The expression level of CCR5in peripheral blood CD4+CD25+CD127-regulatory T rells in gastric cancer patients were evidently higher than that in healthycontrols,and the difference was significan(tt=7.5, P=0.000);(2)The expression levelof CCR5in peripheral blood CD4+CD25+CD127-regulatory T rells was not relatedwith the age of patient(p=0.0.936), sexual distinction(p=0.996), tumor size(p=0.08),but with the histologic grade(p=0.001),clinical stage of the cancer(p=0.003) andlymph node metastasis(p=0.001).(3) The expression level of CCR5in peripheralblood CD4+CD25+CD127-regulatory T rells was positively linear correlated withlymph node metastasis(B=0.783, P=0.000) or pathological stage in gastriccancer(B=0.331,P=0.005), but not with the age of patient, sexual distinction, tumor size and histologic grade.(4) The expression level of CCR5in peripheral bloodCD4+CD25+CD127-regulatory T rells was not correlated with age of patient, sexualdistinction, tumor size, but with pathological stag(er=0.736,P=0.000)or lymph nodemetastasis(B=0.783,P=0.000).Conclusion:The expression level of CCR5in peripheral blood CD4+CD25+CD127-regulatory T rells is up-regulated in gastric cancer patients, is evidently higher thanthat in healthy controls. The expression of CCR5is high correlated with the clinicalstage and lymph node metastasis, point out the higher clinical stage or the morelymph node metastasis,the higher expression level of CCR5in peripheral blood.CCR5and CD4+CD25+CD127-regulatory T rells may participate in the tumorimmunity,may play an important role in the invasion and metastasis of gastric cancer.
