Objective:The dissertation aims to study the expression of TCRαβ/γδ/δ1/δ2 and CXCR4,CCR5 in breast cancer patients' peripheral blood and tumor tissue to analyze their expression features and impacts;and analyze the functional relevance between TCRαβ/γδ/δ1/δ2 and chemokine receptor subgroup CXCR4,CCR5 and the relation between the expression and the disease;and lay the rationale for the experiment and clinical application of tumorous bioimmunity targeted therapy.Methods:Applying flow cytometry FCM to examine the level of TCRαβ,TCRγδ,TCRδ1,TCRδ2,CXCR4,CCR5,cytokine(IL-2,IL-4,IL-5,IL-10,TNF-α,INF-γ),CA125,CA153 and lymphocyte immune function (CD3,CD4,CD8,CD4/CD8,NK,CIK)in 30 breast cancer patients' in peripheral blood before and after operation,and compare with other 30 benign tumors;to examine simultaneously the expressions of normal breast tissues,paracarcionoma breast tissues,and breast cancer tissues TCRαβ,TCRγδ,TCRδ1,TCRδ2,CXCR4,CCR5 in resected sample, together with clinical pathological features to analyze the expression of TCRαβ/γδ/δ1/δ2 and CXCR4,CCR5 in breast cancer patients' peripheral blood and tumor tissues.Results:1.The expressions of 30 breast cancer patients' TCRγδ,TCRδ1,TCRδ2 in peripheral blood are 8.73±7.57%,7.82±5.47%and 6.81±6.32%, respectively,and compare to benign tumors'group,they significantly increased(P=0.000);the expressions of chemokine receptor CCR5,CXCR4 are 26.64±5.46%,41.10±8.08%,and compare to benign tumors'group,they significantly increased(P=0.000);2.Compared with normal breast tissues' expression,the TCRγδ's expression in paracarcinoma breast tissues:6.78±5.48%in normal breast tissues,23.58±14.88%in paracarcinoma breast tissues and 17.30±18.16%in breast cancer tissues and the divergence embodies the statistical signification(P<0.05);and the comparison between but the comparison between paracarcinoma breast tissues and breast cancer tissues has no statistical signification;the comparisons of expressions of TCRγδin normal breast tissues,paracarcinoma breast tissues and breast cancer tissues are 8.39±7.51%in normal breast tissues,23.77±15.00 %in paracarcinoma breast tissues,and 11.65±11.07%in breast cancer tissues.The divergence embodies the statistical signification(P<0.05); comparing to paracarcinoma breast tissues and breast cancer tissues,the divergence embodies the statistical signification(P<0.05);the comparisons of expressions of TCRδ2 in paracarcinoma breast tissues and breast cancer tissues and the expression in normal breast tissues are: 6.09±6.24%in normal breast tissues,18.44±16.35%in paracarcinoma breast tissues,8.16±7.03%in breast cancer tissues,and the divergence embodies the statistical signification(P<0.05);comparing to paracarcinoma breast tissues and breast cancer tissues,the divergence embodies the statistical signification(P<0.05),and the expression of paracarcinoma breast tissues are higher than breast cancer tissues.3.The expressions of normal breast cancer tissues,paracarcinoma breast tissues,CCR5 in breast cancer tissues are 10.00±1.11%,23.63±3.41%,53.34±7.50%;the expressions of CXCR4 are 12.57±2.45%,36.28±6.13 %,67.26±3.86%;these two expressions in tissues show that the breast cancer tissues' are higher than paracarcinoma breast tissues,and paracarcinoma breast tissues' are higher than normal breast tissues'. There are significant change and the divergence embodies the statistical signification(P<0.001);4.The comparison among the expression levels of TCRδ1,TCRδ2 in breast cancer patients'peripheral blood and Tumor primary stove size,lymph node metastasis,tumor markers CA-125 and CA-153 have statistical signification(P<0.05);the comparison of TCRαβ,TCRγδ,TCRδ1,TCRδ2 before and after operation has statistical signification(P<0.05); the comparison between the expression levels and relevance of TCRαβ,TCRγδ,TCRδ1,TCRδ2 in breast cancer tissue andTumor primary stove size,lymph node metastasis,CA-125,CA-153 has no statistical signification(P>0.05);5.The comparisons among the expression levels of CXCR4,CCR5 in breast cancer patients'peripheral blood and breast cancer tissues and the expressions of Tumor primary stove size,lymph node metastasis,tumor markers CA-125 and CA-153 have statistical signification(P<0.05);6.The expression levels of CXCR4,CCR5,TCRδ1,TCRδ2 in breast cancer patients'peripheral blood are related,and the CXCR4,CCR5 is positive correlation(P<0.05);both TCRδ1 and TCRδ2 display chglish (P<0.05);when the expression levels of chemokine receptor CXCR4,CCR5 in breast cancer patients'peripheral blood display high, so as TCRδ1,TCRδ2(P<0.05);the expression levels of breast canser tissues CXCR4,CCR5 are related,when the expression of CXCR4 is high,so as the CCR5(P<0.05);there is positive correlation between CXCR4,CCR5 in breast cancer patients' peripheral blood and CXCR4, CCR5 in breast canser tissues(P<0.05);the expression levels of TCRδ1,TCRδ2 in breast cancer patients' peripheral blood and breast canser tissues are related that both of them display high expressions(P<0.05);the expression levels of TCRγδin breast cancer patients' peripheral blood and in breast cancer tissue is positive correlation(P<0.05).Conclusion:The expressions of TCRγδ,TCRδ1,TCRδ2 and CXCR4,CCR5 in breast cancer patients' peripheral blood are high,and obviously higher than the benign tumor group,and these expressions in breast cancer patients' peripheral blood and breast cancer tissues have a correlation; they are linked to the expressions of Tumor primary stove size,lymph node metastasis,and tumor markers CA-125 and CA-153,the results have the statistical signification;they might be related to the occurrence and metastasis of breast cancer which play a pivotal role in advancing breast cancer's growth and development.That could be the significant index in predicting tumor metastasis and evaluating prognosis,as well as be a new target for cancer treatment and to provide some new clues and ideas to the molecule targeted therapy of breast cancer.
