Objects: To investigate the effects of ulinastatin (UTI) on protection of liver injury induced by CCl4.Methods: The rat model of liver injury was established with CCl4by intragastric administration. A total of50SD rats were randomly divided into five different doses:control group, injury group, low-dose(2.5×104U·kg-1),middle-dose(5×104U·kg-1) and high-dose (1×105U·kg-1). UTI was intravenously. The serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were analyzed by automatic biochemical analyzer. The morphology of liver was observed by hematoxylin-eosinstaining. The expressions of IL-1β, IL-2, IL-6, IL-8, TLR4, NF-κB and TNF-α in liver were detected by RT-PCR and Western Blot.Results:(1) The serum AST and ALT increased significantly in rats induced by CCl4.(2) The expressions of IL-1β, IL-2, IL-6, IL-8, TLR4, NF-κB and TNF-α in liver increased significantly induced by CCl4.(3) UTI can improve morphology of liver in rats after liver injury.(4) The serum AST and ALT decreased significantly in rats with liver injury when treated with UTI in a dose-dependent manner, specially for high dose of UTI.(5) The expressions of IL-1β, IL-2, IL-6, IL-8, TLR4, NF-κB and TNF-α in liver decreased significantly when treated with UTI.Conclusion: UTI have a protective effect on CCl4-induced liver injury in rats in a dose-dependent manner at a certain range of concentrations. The mechanism may be inhibition of expressions of IL-1β, IL-2, IL-6, IL-8, TLR4, NF-κB and TNF-α in liver were inhibited by UTI.
