Objective Study on the health of rat skin fibroblasts cultured in vitro withsodium hydrosulfide (NaHS), L-propargylglycine (PPG) collagenⅠ(CoⅠ) andcollagen Ⅲ (CoⅢ) changes in the expression levels of mRNA.Methods Health in primary rat skin fibroblasts in vitro culture, subculture, selection of fiber cells were plated into third generations, were randomly divided intoblank control group, normal saline control group, L-propargylglycine (PPG) groupand sodium hydrogen sulfide (NaHS) group, each of the3side holes. To besynchronized to the composition of fiber cells after48h incubation, the interventionof agent.24h RNA Trizol extraction, identification, RT-PCR method for thedetermination of collagenⅠ(CoⅠ) and collagen Ⅲ (CoⅢ) gene expression level. Results The RT-PCR method measured the expression of collagenⅠ, Ⅲ mRNAlevels were not significantly different. Conclusions Group NaHS (N1, N2, N3), blank control group, saline controlgroup, PPG group (P1, P2, P3) in the normal rat skin fibroblast collagenⅠ, Ⅲ andmRNA were no significant differences in the level of expression. RT-PCR method formeasuring hydrogen sulfide (H2S) on normal rat fibroblasts secrete collagenⅠ, Ⅲalmost no significant influence. Hydrogen sulfide (H2S) to be further argued on burnwound healing.
