OBJECTIVE:Gastric carcinoma, one of common malignant tumors, severely threatens thehuman health. The research on apoptosis signal transduction pathway becomes oneof research hotspots on apoptosis. Caspase cascade reaction has already beenregarded as the central link of apoptosis process. Caspase-3is at the core location ofprotease cascade reaction. To detect Caspase-3protein expression in tissues such as gastriccancer, para-carcinoma and normal gastric mucosa among those of patients with gastriccarcinoma mainly through immunohistochemistry; to conduct retrospective analysis onexpression of serum tumor markers such as CEA and CA19-9in clinical data of patients and todiscuss the protein expression of tissue Caspase-3of patients with gastric carcinoma as well asthe relevance and clinical significance of occurrence and invasion&metastasis of serum tumormarkers in gastric carcinoma.METHOD:Detect protein expression of Caspase-3in tissues such as gastric carcinoma, para-carcinomaand normal gastric mucosa and conduct retrospective analysis on detection result of serum tumormarkers including CEA and CA19-9in clinical data of patients through immunohistochemistrySP; analyze the relations among the expression change and sex&age of patients with gastriccarcinoma, location, size and general type of tumor as well as many clinical biological behaviors such as differentiation degree, infiltration depth, lymphatic metastasis and clinical stages; allpatients selected are not given chemotherapy and radiotherapy before operation.Statistical method: Analyze all data with SPSS17.0statistical software; check the differencein positive rate among different groups with X2and conduct rank sum test of two-samplecomparison to check ranked data; when P<0.05, it is indicated that there is remarkabledifference.RESULT:1. Expression positive rate of caspase-3in gastric carcinoma reaches50%and thatin normal mucosa tissue reaches71%(x2=6.219, p=0.013). There is no relevance((P>0.05)) between caspase-3protection expression in gastric carcinoma tissue andage, sex, tumor size, infiltration depth and lymphatic metastasis; caspase-3proteinexpression in gastric carcinoma is related to Lauren tissue typing while that inintestinal pattern group is obviously higher than that in diffuse type group (P<0.05);as for the relationship between caspase-3protein expression in gastric carcinoma, theexpression level of high-moderately differentiation group is apparently higher thanthat in low differentiation group (P<0.05); as for the relationship between caspase-3protein expression in gastric carcinoma tissue and that in clinical staging, theexpression level of clinical staging namely I-II stage is apparently higher than that ofIII-IV stage (P<0.05).2. Positive rate of CEA expression in serum of patients with gastric carcinoma is39%; that of CA19-9in serum of patients with gastric carcinoma is50%. There is norelevance between CEA and A19-9in serum of patients with gastric carcinoma andage, sex, tissue differentiation degree and Lauren typing (P>0.05); as for therelationship between CEA and CA19-9in serum of patients with gastric carcinomaand tumor size, the expression percentage of the group larger than3cm is smaller than that smaller than3cm (P<0.05). As for the relationship between CEA andCA19-9in serum of patients with gastric carcinoma and infiltration depth, positiveexpression percentage of s apparently higher than that of tumor in (P<0.05). As forthe relationship between CEA and CA19-9in serum of patients with gastriccarcinoma, the positive expression percentage of those in group with lymphaticmetastasis is apparently higher that of those in group without lymphatic metasisgroup (P<0.05). as for the relationship between CEA and CA19-9in serum ofpatients with gastric carcinoma and clinical staging, the expression level of clinicalstaging namely III-IV stage is apparently higher than that of I-II (P<0.05).3. There is no relevance between caspase-3protein expression in gastric carcinomatissue and the rise in concentration of CEA and CA19-9in serum of gastriccarcinoma.CONCLUSION:1. The expression percentage of caspase-3protein in gastric carcinoma tissue islower than that in normal gastric mucosa. There is remarkable difference between itand gastric carcinoma tumor Lauren typing, differentiation degree of gastriccarcinoma tissue and clinical staging, but there is no remarkable difference betweenit and patient’s age, sex, tumor size, infiltration degree and lymphatic metastasis.2. There is remarkable difference between the rise in concentration of CEA andCA19-9in serum of gastric carcinoma and tumor size, infiltration degree, lymphaticmetastasis and clinical staging, but there is no remarkable difference between it andpatient’s age, sex, Lauren typing and differentiation degree of tumor tissue.3. There is no relevance between caspase-3protein expression in gastric carcinomatissue and the rise in concentration of CEA and CA19-9in serum of gastriccarcinoma.
