Expression And Significance Of Puma And Caspase-9in Gastric Carcinoma

Objective:To investigate the expression of Puma and Caspase-9in difference gastric mucosal lesions and the relationship between them. And explore the effects of Puma and Caspase-9on the occurrence and development of gastric carcinoma.Methods:The SABC immunohistochemical method was used to detect the expression of Puma and Caspase-9in90different gastric mucosal lesion. Results were analyzed by SPSS version13.0.Results:The expression of Puma and Caspase-9in different gastric mucosal lesion:(1). The positive expression of Puma in chronic superficial gastritis, chronic atrophic gastritis, intestinal metaplasia, atypicalhyperp-lasia, gastric carcinoma was75.00%,60.00%,53.00%,33.33%,22.86%, respectively, which demonstrated the decreasing tendency. The positive expression of Puma protein between chronic superficial gastritis and gastric carcinoma, chronic atrophic gastritis and gastric carcinoma, intestinal metaplasia and intestinal metaplasia, which was significantly different(p<0.05).(2) The positive expression of Caspase-9in chronic superficial gastritis, chronic atrophic gastritis, intestinal metaplasia, atypicalhyperplasia, gastric carcinoma was91.67%,80.00%,60.00%,44.44%,34.29%, respectively, which demonstrated the decreasing tendency. The positive expression of Caspase-9protein between chronic superficial gastritis and gastric carcinoma, chronic atrophic gastritis and gastric carcinoma, which was significantly different(p<0.05).3. The positive expression of Puma was correlated to clinical staging, lymph node metastasis and tissue differentiation (p<0.05), there was no correlation to patients’gender, age and invasive depth (p>0.05). The positive expression of caspase-9was correlated to lymph node metastasis (p<0.05),there was no correlation with patients’gender, age, clinical staging, tissue differentiation and invasive depth(p>0.05).The Spearman correlation analysis suggested that the expression of puma and caspase-9was positively correlated (r=0.467, p <0.05)Conclusions:Low expression of Puma and Caspase-9in gastric cancer showed that gastric cancer was correlated with apoptosis. We extrapolated that the suppression of Puma activating Caspase-9plastochondria channel possibly took part in the gastric carcinoma occurrence. The combination of the expression of puma and caspase-9may contribute to the differential diagnosis of gastric cancer.
