Parkinson’s disease (PD) is a disease of the central nervous system that leads tosever difficulties with body motions.Typical symptoms of PD include tremor, rigidity,slowed body movements, unstable posture and difficulty in walking. The experimentis aim to study the effect and mechanisms of Danshensu on motor disorders in mice ofParkinson’s disease.The methods we use are show as following: Thirty two C57/BL male brownmice were randomly divided into four groups: control group; MPTP model group;Danshensu group (15mg/kg); Danshensu group (60mg/kg). Danshensu groups wereinjected with Danshensu for14days, dose amounted to60mg/kg and15mg/kg dailyrespectively. The mice were induced by l-Methyl-4-phenyl-l,2,3,6-TetrahydropyridineHydrochloride (MPTP-HC1,30mg/kg daily for7days) from the8th day of the stuty.One hour after the last administration the effect of Danshensu on the motor behaviorin PD mice is evaluated. The pole test, rotorod test and the open field test are applied.The contents of dopamine (DA) and serotonin (5-HT) in the striatum are detected byusing high-performance liquid chromatography (HPLC). Immunohistochemicalmethod is employed to detect the numbers of tyrosine hydorxylase (TH) positiveneurons in the substantia nigra in PD mice. And the levels of TNF-α and IL-1β wereassayed by ELISA.The results are shown as following:1, Compared with control group, the poletest time of MPTP model group was significantly longer and the rotorod time, thetotal distance and average velocity in open field test was significantly shorter.Compared with model group, the pole test time of Danshensu groups weresignificantly reduced, and the rotorod time, the total distance and average velocity inopen field test were significantly increased.2, Compared with control group, thecontents of DA and5-HT in the striatum of the MPTP model group were significantlyreduced, the contents of DA and5-HT level of the Danshensu group weresignificantly increased.3, Compared with control group, Positive neurons of Tyrosinehydorxylase (TH) in substantia nigra pars compacta was significantly reduced, while Danshensu group was significantly increased.4, Compared with control group, theTNF-α and IL-1β level in the striatum of MPTP model group were significantlyincreased. Compared with MPTP model group, the TNF-α and IL-1β level in thestriatum of Danshensu were significantly reduced.The conclusions we reach are that Danshensu can significantly improve themotor behavior of PD mice in part by reducing the expression of TNF-α and IL-1βand the protective effects on DA nerve cell inducing to increasing the contents of DAand5-HT.
