Objective:(1)Through a prospectively designed algorithm was applied retrospectively, analyzing tothe transcriptome molecular characteristics of maternal peripheral blood nucleated cell inspontaneous premature birth.(2) Based on the mRNA expression profile of premature labor, differentially expressedgenes were selected and may be contribute to explore the moleclar mechanism of spontaneouspreterm birth.Materials and methods:(1)The mRNA expression profile: the mRNA expression profile of21initial samples wasinvolved in this study, including10SPTB and11TB. The gene expression features of SPTBgroup and TB group were analyzed, then integrative analysis was performed using the data.(2)Real time-PCR (RT-PCR): according to the gene expression profiles, we selected13up-regulated different genes for further validation in47independent samples (21SPTB and26TB) by Real time-PCR.Results:(1) The reslts of the mRNA expression profile: the results revealed that up-reglated geneswith the number of241in biological process referred to Inflammation, immune response, cellproliferation, metabolism of DNA replication, DNA metabolism, RNA shear, RNA metabolismand chromatin assembly, etc; the down-reglated genes with the number of690were associatedwith steady state, signal transduction, ion transport and nucleic acid catabolism, etc.(2) The results of Real time-PCR: The results revealed the13genes differentially expressedin47independent samples,The levels of NF1in PB group is apparent remarkable higher thanSPTB group(P<0.05,P=0.008), the differences were significant; the levels of CAMK2D inSPTB group is apparent remarkable higher than TB group(P<0.05,P=0.028), the differenceswere significant. Conclusions:(1) The occurrence process of spontaneous preterm birth may be relative to re-activation orre-inhibition of some genes.(2) NF1and CAMK2D gene abnormal expression may be involved in physiopathologychanges of premature birth, which may be candidate molecular markers for prediction ofspontaneous preterm birth.
