Objective:To detect the level expression of KLK9gene in the tissues of liver cancer, andanalyse the clinical significance and relationship between the express of KLK9geneand the clinicopathologic features of liver cancer(including patient’s age, sexuality,the size of specimen, the degree malignant of liver cancer, TNM stage, whether thetransfer and HBsAg positive).Investigate the relationship between the expression ofKLK9gene and the malignance degree of cell by means of detecting the expressionof KLK9gene in a normal liver cell line and four different malignant degree ofhepatocarcinoma cell lines. To study the roles of KLK9and related molecularmechanism in the occurrence and development of liver cancer, and provide thetheory and data base for the diagnosis and biological target therapy of liver cancer.Methods:1、To collect50patient’s liver cancer tissues,all of the patient had completeclinical data(no other history of malignant tumors, radiotherapy and chemotherapybefore operation)and from the edge of the lesion2cm cut out part of the normal livertissues as an paracarcinoma control group, and25cases of hepatolithiasis andtraumatic removal of part of normal liver tissue as a normal control group.2、 To test the expression of KLK9protein in the liver cancer tissues(n=50),normal tissues(n=25) and paracarcinoma tissues by means of immunohis-tochemistry. To analyse the relationship between the expression of KLK9protein andclinicopathologic features of the liver cancers.3、To detect the expression of KLK9mRNA in the liver cancer tissues,normaltissues and paracarcinoma tissues by means of semiquantitative RT-PCR,andinvestigate the relationship between the level expression of KLK9mRNA andclinicopathological features of the liver cancers.4、We had detected the differential expression of KLK9mRNA and protein in a normal liver cell line and four liver cancer cell lines with different degreemalignance by way of RT-PCR and Western Blot,and analysed the relationshipbetween the level expression of KLK9mRNA and protein and the degreemalignance of liver cancer cell.Results:1、The rate expression of KLK9gene mRNA and protein in the liver cancercells was significantly higher than in paracarcinoma tissues and normal liver tissues(P<0.05).2、The expression of KLK9mRNA and protein was related to the metastasisand the degree of malignance and clinical staging of the liver cancer and the sex ofpatients(P≤0.05),but wasn’t associated with age, and HBsAg of patients and thesize of tumors (P>0.05).3、The expression of KLK9mRNA and protein in the liver cancers wereaccompanied with the degree malignance of the liver cancer cells, the higher degreemalignance,the higher level expression of KLK9gene mRNA and protein,but thelevel expression of KLK9in normal was low.Conclusions1、The expression level of KLK9gene protein and mRNA in liver cancer tissuesare related with the degree malignance of the liver cancer, the expression level ofKLK9in the normal liver tissues are low,but the expression level of KLK9in livercancers are high, and the higher degree malignance of the liver cancer, theexpression level of KLK9are higher.2、The expression level of KLK9gene is related with the distant metastasis ofliver cancer cells, the expression level of KLK9in liver cancer tissues withoutdistant metastasis, the expression level of KLK9in liver cancer tissues with distantmetastasis are higher.3、The expression of KLK9gene in liver cancer tissues are related with the sexof patients,the expression level of KLK9gene in male patients are higher in femalepatients. 4、The expression of KLK9gene in liver cancer cell lines have to do with thedegree malignance of the liver cancer cell lines, the expression level of KLK9genein normal liver cell lines are lower in four liver cell lines, and the higher degreemalignance of the liver cancer cell lines, the expression level of KLK9are higher.
