The polyphenol content is relatively in tea beverages, and the flavonoids largelypresent in tea polyphenols, such as catchins, theaflavins and thearubigins, which havephysiological effects by scavenging free radicals that are generated by metabolic pathwaysin body tissue or introduced by foods, drugs, and environmental in pollutants. Teapolyphenol is considered to be the natural antioxidant due to its function of oxidationresistance and inhibiting the lipid peroxidation. Because of differences of varieties,climate environment, sunshine and manufacturing, the types and contents of polyphenolsin tea were impossible completely same. Hence, the effect of antioxidant activity andpharmacological functions is not the same.For the problems such as, the substance construction of polyphenols in theHuangshan Maofeng and Keemun black tea are not clear, the effect on the protection of liver is also not clear, hence, HPLC analysis method of tea polyphenols on the basis ofseparation and preparation HMTP and KBTP is established in this research, define thecomposition and content of the two kinds of tea polyphenols, explore anti-oxidativedamage of Huangshan Maofeng and Keemun black tea polyphenols with the organiccombination of in vitro and in vivo study, explain pharmacological actions of theirresistance to liver damage from the level of the whole animal, provide experiment basisfor further development of Huangshan Maofeng tea and Keemun black tea.In this research, the tea polyphenols were extracted from these two kinds of tea,respectively. Then we successfully separate the tea polyphenols which extracted from thetwo kinds of tea and determine the content of tea polyphenolic compounds for the firsttime. This study established HPLC analysis method for the separation and analysis of the8kinds of HMTP and the6kinds of KBTP, identify the composition of HMTP and.KBTP.And showed that three major polyphenolic compounds were respectively presented inHMTP, were epigallocatechin (271.2μg/mg), followed by rutin (239.3μg/mg) and EC(89.3μg/mg).. And in KBTP were EC(439.82μg/mg)、rutin(125.65μg/mg) and EGC(66.81μg/mg).The study of the antioxidation activities show both HMTP and KBTP have strongscavenging activities against DPPH, O2, and OH, and ferric-reducing antioxidantpowers (FRAP) in vitro, compared with the positive drug group (BHT), and there isdose-effect relation.Administration of HMTP at200,400and800mg/kg·bw in mice prior to CCl4injurysignificantly decreased the CCl4-induced elevation of serum ALT, AST and ALP activities,and prevented the increase in hepatic MDA levels, and the activities of GSH-Px and SODin hepatic tissue are improved at the same time (p<0.05), relative to CCl4-intoxicated mice.Liver pathological observation also confirmed the protection on CCl4-caused histologicalalteration, especially in the high dose groups, mutation cells remarkably descreased, andthe shape of liver cell are in good condition, compared with the CCl4-intoxicated group.These results indicate that both HMTP and KBTP have protective effects againstCCl4-induced liver injury in mice. To sum up, this study establish HPLC analysis method for the separation and analysisof the10kinds of polyphenols, identify the composition of HMTP and KBTP, then adoptexperiments of DPPH、O2、OH scavenging ability and reducing power of Fe3+, theresults show these polyphenols have antioxidant activity in vitro, finally build the modelof animal’s CCl4induced liver injury to evaluate the effects of the anti-oxidation damageof the two tea polyphenols. This line of research will lay the foundation for antioxidanteffect study of Huangshan Maofeng green tea and Keemun black tea.
