Lung cancer is one of the most common tumor in the world, its morbidity andmortality in men ranked first, in women also ranked second only to breast cancer. Withthe changes of lifestyle、aging of population、environmental pollution, the incidence oflung cancer is still increasing all over the world,and particularly in the developingcountries. Liver, brain,bone is the most frequently in lung cancer. The distant metastasis isthe main reason leading to the decreaseing of quality of life of the patients and poorprognosis.IQGAP1proteins have been identified in various organisms, ranging from yeast tohumans. IQGAP1, which participates in numerous signaling pathways.IQGAP1achieve these effects through combining with and regulating the function of manyinteracting proteins,such as E-cadherin, Ca2+/Camlodulin,S100B, Cdc42/Rac1.There are alarge number of reported that IQGAP1was overexpression in different tumor tissues andcell lines.However, the mechanism underlying lung caner remains unclear and rarereported.Purose:(1) The objective of this study was to acknowledge the diversity expression ofIQGAP1in non-small cell lung cancer; (2) To study the effect of IQGAP1on the behavior of the no-small cell lung cancer.Methods:(1) The expression of IQGAP1inA549, PC14, PC14/B were examined by Real-timePCR and Western blot respectively;(2) The expression of IQGAP1was down-regulated by RNAinterence (RNAi) inPC14/B and identify by RT-PCR and Western-blot. In colony-forming assay,viroscratch test and transwell based methods were used to detect the effect of IQGAP1on the proliferation, migration and invasion ability of no-small cell lung cancercells;(3) The expression change of E-cadherin in PC14/B,NC-PC14/B, IQGAP1RNAi-PC14/B was analyzed by Western blot and immunofluresenceResults:(1) IQGAP1was over-expressed in non small cell lung cancer PC14/B;(2) The efficient transfection was measured by real time-PCR and Western blot inPC14/B. Down regulation of IQGAP1can decrease proliferation、migration andinvasion potential in PC14/B cell lines;(3) E-cadherin was partly involved in the IQGAP1in the lung cancer.Conclusion:IQGAP1is a multifunctional oncogene, widely involved in cell adhesion, cellmigration and so on. IQGAP1has different expression in vary non small cell lung cancer.Compared to A549、PC14, IQGAP1with high expression in PC14/B which is specificbrain metastatic cell lines, and can enhance cell proliferation, invasion,inhibit theexpression of E-cadherin.
