Nervilia Herb is the tuber and the whole plant of Nervilia forddi (Hance) Schltr. Ithas been used as a traditional Chinese medicine for the treatment of various diseases,such as tuberculosis, cough, hemoptysis, crewels, pyogenic infections, traumaticinjury, infantile malnutrition bronchitis, scrofula, pyogenic infections and pneumonia.Thirteen compounds were isolated from petroleum ether fraction from thealcoholic extract of Nerviliae fordii (Hance) Schltr and eleven compounds of themwere identified on the basis of spectral and physicochemical analysis. Their structureswere established as β-sitosterol (1), stigmaterol (2),(9R,10R,7E)-6,9,10-trihydroxyoctadec-7-enoic acid (3),(20S,22E,24R)-ergosta-7,22-dien-3β,5α,6β-triol(4), stigmast-22-en-3β,6β,9β-triol (5), ergosta-7,22-dien-3β,5α,6α-triol (6),3β-hydroxystigmasr-5,22-dien-7-one (7),3β-hydroxystigmasr-5-en-7-one (8),aurantiamide acetate (9), aurantiamide benzoate (10) and3β,5α,9α-trihydroxyergosta-7,22-dien-6-one (11). Compounds3,5~8,10and11were obtainedfrom the plant for the first time.This paper establishes the fingerprints of crude drug material (Nervilia Herba)from Nervilia fordii by HPLC base on the above study and other research in ourlaboratory on the other parts of Nerviliae fordii (Hance) Schltr. HPLCchromatographic condition are: Welch Materials XB-C18(250mm×4.6mm,5μm)column was used, and the acetonitrile-water (0.03%trifluoroacetic acid) was chosenas the mobile phase in a gradient mode, the flow rate of1.0mL/min, the detectionwavelength was at210nm and the column temperature at35℃. There were twelvecharacteristic peaks indicated in fingerprints and among of them4peaks have theirchemical structures. This method is simple and accurate with a good reproducibility. Itprovides a reference standard for the quality control of the materials from Nerviliaefordii.The Pterospermum Radix comes from the dry root of Pterospermumheterophyllum Hance, and its the other name is banfenghe. Pterospermumheterophyllum Hance is distributed in Guangdong, north Guangxi, Hainan, south Jiangxi, and Guizhou. It has been used in Chinese folk medicine for the treatment ofarthritis, back and leg pain, hand and foot numbness, lumbar muscle strain,hemiplegia and limb paralysis.This paper also establishes the method of fingerprint analysis on crude drugPterospermum Radix from Pterospermum heterophyllum by HPLC and found tencommon peaks. HPLC chromatographic conditions are: Welch Materials XB-C18(250×4.6mm,5μm) column, the acetonitrile-water (0.03%trifluoroacetic acid) asthe mobile phase in a gradient mode, the flow rate of1.0mL/min, the detectionwavelength at205nm, and the column temperature at20℃. The clustering analysisand semblance evaluation were carried out for the analysis of methodology andquality evaluation of the crude drugs. This method is stability, precision andreproducibility, providing a credible method and evidence of basic research for thequality control of Pterospermum Radix.
