Objective This study is to investigate the relationships between protein tyrosinephosphatase non-receptor type22(PTPN22) gene polymorphism at1858C> T and1123G> C and incidences of rheumatoid arthritis (RA) in Tibetan population ofQinghai province, China. Methods87cases of sporadic RA patients in Tibetanpopulation of Qinghai and90cases of healthy individuals were analyzed by SNaPshottechnology at PTPN221858genotypes as well as PTPN221123genotype. Resultswere statistically analyzed using the SPSS16.0software. Means of two samples werecompared by T test while the independence of two samples was examined using χ2test. The significant level was defined as p <0.05for the two-sided test.Results Results at PTPN221858C> T genotype: All87cases in RA group were CCgenotype, while in90cases of control group,89cases were CC genotype and onlyone case was CT genotype.Results of PTPN221858C> T genotype: distribution oftow genotypes between control and RA groups was not significantly different by thechi-square test (X~2=0.97, P=1.00).Results at PTPN221858C> T genotype:therewere three genotypes (GG, GC and CC) existed in the Tibetan population of Qinghaiprovince. RA group: GG7(8%) GC34(39.1%) CC46(52.9%),control group: GG4(4.4%) GC44(48.9%) CC42(46.7%).Results of PTPN221123G> C genotype:distribution of three genotypes between control and RA groups was not significantlydifferent by the chi-square test (X~2=2.232, P=0.328). The gene frequencydistributions of healthy individuals in the Tibetan populations of Qinghai weresignificantly different from those of healthy individuals in Japan (X~2=37.288, P=0.000) and those of Han populations in Shanghai, China (X~2=48.234, P=0.000) aswell as those of Han populations in Hebei, China (X~2=67.181, P=0.000)(Table3).Each genotype in RA group was not significantly different from each other onanti-CCP antibody test.(X~2=0.801, P=0.723).Conclusion PTPN221858C> T genepolymorphism in the Tibetan populations in Qinghai of China is not correlated toincidences of RA;PTPN221123G> C gene polymorphism in the Tibetan populationsin Qinghai of China is probably not associated with incidences of RA;Compared to the healthy populations in Japan, Han populations in Shanghai andHebei of China, gene frequency distributions at PTPN221123of Tibetan healthypopulation of Qinghai of China showed significant differences;PTPN221123genotype of Tibetan populations of Qinghai in RA group was not related to anti-CCPantibodies.
