Obejective:To observe the expression of the BMP2/TGFP3modified plasmid in BMSCs and the effects on proliferationMethods:Bone marrow fluid, which was collected through the adherent culture method,was applied for the primary culture and subculture. The second generation BMSCs were selected for the next-step transfection. pIRES/BMP-2, pIRES/TGFβ3,pIRES/BMP2-TGFβ3and the pIRES plasmid was transfected into the BMSCs respectively. The expression of BMP2and TGFβ3was detected and compared by RT-PCR two days after the transfection. Drow the growth curve, calculate the doubling generation time, then make the comparison of proliferation between each group.Results:The expression of BMP2and TGF-β3mRNA of pIRES/BMP2-TGFβ3group was significantly higher than other groups, BMP2and TGF β3was proved to obtain the stabile expression in the BMCs in double gene groups; doubling generation time of the double gene group was conspicuous shorter than other group.Conclusion:BMP2and TGF β3is expressed stable in the BMCs transfected by the pIRES/BMP2-TGFβ3. The double gene have the synergistic effect in the proliferation of the BMCs. |
